目的:观察苦参碱对人鼻咽癌CNE2细胞裸鼠移植瘤生长及凋亡相关基因表达的影响。方法:20只裸鼠皮下接种CNE2,建立移植瘤模型,成瘤后,随机分成4组,每组5只。苦参碱高、低剂量组分别腹腔注射100、50 mg/kg苦参碱,阳性对照组腹腔注射顺铂2 mg/kg,阴性对照组给予生理盐水。给药7次后,应用体内抑瘤实验测定抑瘤率,分别采用real-time PCR法和Western blot法检测移植瘤组织中bax、bcl-2、caspase-3 mRNA和蛋白的表达。结果:阳性对照组和苦参碱低、高剂量组抑瘤率分别为51.6%和26.4%、43.2%。4组间移植瘤组织中bax、bcl-2、caspase-3的mRNA和蛋白表达差异有统计学意义(F=3.751、16.113、3.918和26.500、9.771、12.255,P<0.05);与阴性对照组比较,阳性对照组和苦参碱高、低剂量组bax mRNA 和蛋白表达及阳性对照组、苦参碱高剂量组caspase-3 mRNA和蛋白表达上调(P<0.05),bcl-2 mRNA和蛋白表达下调(P<0.05)。结论:苦参碱可通过调节CNE2移植瘤组织中凋亡相关基因的表达抑制肿瘤的生长。

목적:관찰고삼감대인비인암CNE2세포라서이식류생장급조망상관기인표체적영향。방법:20지라서피하접충CNE2,건립이식류모형,성류후,수궤분성4조,매조5지。고삼감고、저제량조분별복강주사100、50 mg/kg고삼감,양성대조조복강주사순박2 mg/kg,음성대조조급여생리염수。급약7차후,응용체내억류실험측정억류솔,분별채용real-time PCR법화Western blot법검측이식류조직중bax、bcl-2、caspase-3 mRNA화단백적표체。결과:양성대조조화고삼감저、고제량조억류솔분별위51.6%화26.4%、43.2%。4조간이식류조직중bax、bcl-2、caspase-3적mRNA화단백표체차이유통계학의의(F=3.751、16.113、3.918화26.500、9.771、12.255,P<0.05);여음성대조조비교,양성대조조화고삼감고、저제량조bax mRNA 화단백표체급양성대조조、고삼감고제량조caspase-3 mRNA화단백표체상조(P<0.05),bcl-2 mRNA화단백표체하조(P<0.05)。결론:고삼감가통과조절CNE2이식류조직중조망상관기인적표체억제종류적생장。
Aim:To investigate the effect of matrine on expressions of apoptosis-related genes and growth of human na-sopharyngeal carcinoma CNE 2 cell xenografts in nude mice .Methods:A total of 20 nude mice were subcutaneously inocu-lated with CNE2.After the successful establishment of xenograft model ,the 20 nude mice were randomly divided into 4 groups, 5 rats in each group .Low-and high-dose matrine groups were intraperitoneally injected with matrine at dose of 50 and 100 mg/kg, positive control group was injected with cisplatin ,and negative control group received normal saline .After 7 times administration , the tumor inhibition rate was detected , and the mRNA and protein expressions of bax , bcl-2 and caspase-3 were detected by real-time PCR and Western blot,respectively.Results: The tumor inhibition rates of positive control group, and low-,high-dose matrine groups were 51.6%, 26.4%, and 43.2%;the mRNA and protein expressions of bax,bcl-2 and caspase-3 in xenograft tissue had significant differences (F=3.751,16.113,3.918 and 26.500,9.771, 12.255, P<0.05).Compared with the negative control group , the mRNA and protein expressions of bax in positive con-trol group and low-,high-dose matrine groups and those of caspase-3 in positive control group and high-dose matrine group increased(P<0.05),but those of bcl-2 decreased(P<0.05).Conclusion: Matrine could inhibit the growth of CNE2 xenograft through regulating the expressions of apoptosis-related genes .
