目的:探讨蛛网膜下腔出血(SAH)大鼠血浆及脑脊液神经肽Y(NPY)的变化趋势,分析NPY浓度与脑血管痉挛(CVS)的相关性。方法 SD大鼠25只,随机分为对照组和SAH后1h、12h、1d、7d组共5组,每组5只。除对照组外,另4组枕大池注血制备SAH大鼠模型。放射免疫法测定各组NPY浓度,激光多普勒血流仪测量局部脑血流量(rCBF)。结果 SAH大鼠1h、12h、1d组血液NPY 浓度分别为(44.90依5.92)、(66.27依7.04)、(91.43依10.90)ng/mL,脑脊液 NPY 浓度分别为(102.98依13.54)、(142.38依36.79)、(195.26依22.30)ng/mL,与对照组比较差异均有统计学意义(P<0.05)。Pearson相关分析显示,SAH大鼠血液(r=-0.585,P=0.007)、脑脊液(r=-0.545,P=0.013)NPY含量均与rCBF呈显著负相关。结论 SAH大鼠血液及脑脊液中NPY含量于1h即开始升高,12h达高峰,之后逐渐下降,至7天恢复正常。SAH大鼠血液及脑脊液NPY含量与rCBF呈显著负相关,与CVS存在相关性。

목적:탐토주망막하강출혈(SAH)대서혈장급뇌척액신경태Y(NPY)적변화추세,분석NPY농도여뇌혈관경련(CVS)적상관성。방법 SD대서25지,수궤분위대조조화SAH후1h、12h、1d、7d조공5조,매조5지。제대조조외,령4조침대지주혈제비SAH대서모형。방사면역법측정각조NPY농도,격광다보륵혈류의측량국부뇌혈류량(rCBF)。결과 SAH대서1h、12h、1d조혈액NPY 농도분별위(44.90의5.92)、(66.27의7.04)、(91.43의10.90)ng/mL,뇌척액 NPY 농도분별위(102.98의13.54)、(142.38의36.79)、(195.26의22.30)ng/mL,여대조조비교차이균유통계학의의(P<0.05)。Pearson상관분석현시,SAH대서혈액(r=-0.585,P=0.007)、뇌척액(r=-0.545,P=0.013)NPY함량균여rCBF정현저부상관。결론 SAH대서혈액급뇌척액중NPY함량우1h즉개시승고,12h체고봉,지후축점하강,지7천회복정상。SAH대서혈액급뇌척액NPY함량여rCBF정현저부상관,여CVS존재상관성。
Objective To observe the changes of neuropeptide Y (NPY) in sera and cerebral fluid (CBF) in subarachnoid hemorrhage (SAH) rats and its relationship with cerebral vasospasm. Methods Twenty-five SD rats were randomly divided into control group, post-SAH 1h, 12h, 1d, and 7d groups (n=5). Except for control group, SAH model was established by injection of autologous blood into the rat's cisterna magna. The content of NPY was determined by radioimmunoassay. Regional cerebral fluid was measured by laser Doppler flowmeter. Results The contents of serum NPY were(44.90±5.92), (66.27±7.04), and(91.43±10.90)ng/mL and the contents of NPY in CBF were(102.98±13.54), (142.38±36.79), and(195.26±22.30)ng/mL in post SAH 1h, 12h, 1d groups, with significant differences when compared with control group (P<0.05). Pearson correlation analysis showed that the contents of NPY in sera and CBF were negatively correlated to regional CBF(r=-0.585,P=0.007; r=-0.545,P=0.013). Conclu-sion The contents of NPY in sera and CBF began to increase 1h after SAH, peaked at 12h, and reduced to baseline at 7d. It is negatively correlated to regional CBF, indicating a relationship with cerebral vasospasm.
