为寻找操作简单、分析准确度高、速度快的甜茶多酚的分析方法,以没食子酸为对照品,对测定甜茶中总多酚含量的检测波长、碳酸钠溶液用量、Folin-Ciocalteu 试剂用量、反应温度及时间对吸光值的影响进行了研究。结果表明:Folin-Ciocalteu 比色法测定甜茶总多酚的最佳反应条件为 FC 试剂2 mL,15%碳酸钠溶液7.5 mL,在30℃下反应60 min,该条件下的平均回收率为100.23%(RSD=0.45%)。该方法精确度较高,准确性好,可用于甜茶中总多酚含量的测定。

To set up an assay method for the determination of total polyphenol in sweet tea,the determination conditions of total polyphenol content in sweet tea including the detection wavelength,the amount of sodium carbonate solution,Folin-Ciocalteu reagent,reaction temperature and time were studied and gallic acid was used as a standard.The results indicated that the optimum conditions for determining total polyphenol in sweet tea were as follows:2 mL FC reagent,7.5 mL 15% sodium carbonate solution, treating at 30℃ for 60 min,the average recovery was 100.23%(RSD= 0.45%).This method was simple and quick,the result was accurate,repeatable and stable.It can be used for the determination of total polyphenol content in sweet tea.
