为了获得较高的蝉花核苷碱基,为其品质评估提供方法,采用超声波法对蝉花中的核苷和碱基进行提取,通过单因素和正交试验确定最优提取条件,运用高效液相色谱法对提取液中的8种核苷碱基类物质进行分析测定。结果表明:最优提取条件是以纯水为提取溶剂,料液比1∶40,提取液 pH 4,超声提取温度60℃,提取90 min,在该条件下的总核苷和碱基的提取得率可达2.74%。

In order to gain more nucleosides and bases from C.ciecadae and provide a method to evaluate its quality,the nucleosides and bases were extracted from C.ciecadae by ultrasonic method, which was optimized through a single-factor test and an orthogonal test.Then eight kinds of nucleosides and bases extracted from C.ciecadae were analyzed and detected by HPLC method.The results showed that the optimum extraction conditions included pure water as the solvent,solid-liquid ratio 1:40,pH 4, extraction temperature 60℃ and extraction time 90 min.Under these conditions,the extraction rate of total nucleosides and bases could reach 2.74%.
