目的:探讨葛根素对氧化应激状态下人视网膜色素上皮细胞( ARPE-19 cells )的保护作用。方法用200μmol/L过氧化氢( H2 O2)诱导氧化应激,不同浓度葛根素作用于ARPE-19细胞24 h。应用比色法观察每组中超氧化物歧化酶( SOD)、谷胱甘肽( GSH)及丙二醛( MDA)含量的变化。结果H2 O2能够降低SOD活性,降低GSH含量,并且增加MDA含量,应用葛根素剂量依赖性地减少了以上效应。结论葛根素能够通过调节细胞内抗氧化标记物,保护人视网膜色素上皮细胞抵御H2 O2诱导的氧化应激。

목적:탐토갈근소대양화응격상태하인시망막색소상피세포( ARPE-19 cells )적보호작용。방법용200μmol/L과양화경( H2 O2)유도양화응격,불동농도갈근소작용우ARPE-19세포24 h。응용비색법관찰매조중초양화물기화매( SOD)、곡광감태( GSH)급병이철( MDA)함량적변화。결과H2 O2능구강저SOD활성,강저GSH함량,병차증가MDA함량,응용갈근소제량의뢰성지감소료이상효응。결론갈근소능구통과조절세포내항양화표기물,보호인시망막색소상피세포저어H2 O2유도적양화응격。
Objective To investigate the protective effect of puerarin on ARPE-19 cells against oxidative stress.Methods ARPE-19 cells exposed to 200μmol/L hydrogen peroxide (H2O2) were incubated with various concentrations of puerarin for 24 hours.The activity of the superox-ide dismutase ( SOD) , and the change of the content of glutathion ( GSH) and malonaldehyde ( MDA) in each group were observed by chromatometry .Results H2 O2 decreased the activity of SOD, decreased the content of GSH and increased the content of MDA , that were reduced with different concentrations of puerarin in a dose-dependent manner .Conclusion Puerarin could protect ARPE-19 cells against H 2 O2-induced oxidative stress through modulation of intra-cellular antioxidant biomarkers .
