以口岸经常截获的红脂大小蠹、黄杉大小蠹、山松大小蠹、间大小蠹、红翅大小蠹和落叶松大小蠹等6种大小蠹属昆虫为样本,根据其昆虫检索表制作了近缘关系树;利用分子手段对其线粒体COI基因进行扩增,分析同源序列的多样性、遗传距离和系统进化关系,并将近缘关系树和分子进化树进行比较,研究利用分子手段对大小蠹属昆虫进行快速鉴定的可行性和准确性。结果表明:同种的大小蠹碱基差异较小,而不同种间的碱基差异较大,能明显区分出不同种;大小蠹属的遗传距离和邻接法系统进化树也证实,分子鉴定结果和检索表分类的结果一致。可利用分子鉴定的方法对不同种的大小蠹属昆虫进行快速准确的鉴定。

Taking Dendrotonus valens,D.pseudotsug,D.ponderosae,D.adjunctus,D.rufipennis and D.simplex 6 Dendrotonus species often intercepted in ports as the smaples,this paper constructed a Close Sib Relationship Tree;it used molecule measures to amplify mitochondrial COI genes,it analysed the relationships among same source sequence diversity,heredity distance and system evolution, and Close Sib Relationship Tree and Molecule Evolution Tree were compared,and it researched the feasibility and accuracy of Dend-rotonus insects rapid identification with the molecular techniques.The results showed that the homogeneous Dendrotonus bases had the small difference and different kinds of it had the larger difference,the different species could be clearly distinguished;the Dendro-tonus genetic distance and Neighbour-Joining System Evolution Tree also confirmed that the molecular identification results and the retrieval table classification results were the same.The molecular identification methods were used to do the fast accurate identifica-tion for different kinds of Dendrotonus insects.
