目的:观察5-氟尿嘧啶(5-FU)联合DC-CIK细胞对胃癌BGC-823细胞及其侧群(side population,SP)细胞的体外杀伤效应。方法从胃癌BGC-823细胞中分离获得BGC-823-SP细胞,将BGC-823细胞设为A组,BGC-823-SP细胞设为B组;取健康人外周血单核细胞,培养DC-CIK细胞;用MTT比色法测定5-FU、DC-CIK细胞对A、B 两组胃癌细胞的杀伤率,并测定两者联合对A、B两组胃癌细胞的杀伤率。结果①不同浓度5-FU对A组胃癌细胞杀伤率均高于B组,即B组比A组对5-FU的耐药性更强;②不同效靶比DC-CIK细胞对两组细胞均有较强杀伤作用,且随着效靶比的增加杀伤效果逐渐增强。同一效靶比下,DC-CIK细胞对A组与B组胃癌细胞的杀伤率比较,差异均无统计学意义( P >0.05);③5-FU联合DC-CIK细胞对两组胃癌细胞的杀伤率均明显高于单独应用5-FU或DC-CIK细胞。结论5-FU联合DC-CIK细胞能够显著抑制胃癌BGC-823细胞及其BGC-823-SP细胞的生长,可为临床化疗联合生物免疫治疗胃癌提供理论依据。

목적:관찰5-불뇨밀정(5-FU)연합DC-CIK세포대위암BGC-823세포급기측군(side population,SP)세포적체외살상효응。방법종위암BGC-823세포중분리획득BGC-823-SP세포,장BGC-823세포설위A조,BGC-823-SP세포설위B조;취건강인외주혈단핵세포,배양DC-CIK세포;용MTT비색법측정5-FU、DC-CIK세포대A、B 량조위암세포적살상솔,병측정량자연합대A、B량조위암세포적살상솔。결과①불동농도5-FU대A조위암세포살상솔균고우B조,즉B조비A조대5-FU적내약성경강;②불동효파비DC-CIK세포대량조세포균유교강살상작용,차수착효파비적증가살상효과축점증강。동일효파비하,DC-CIK세포대A조여B조위암세포적살상솔비교,차이균무통계학의의( P >0.05);③5-FU연합DC-CIK세포대량조위암세포적살상솔균명현고우단독응용5-FU혹DC-CIK세포。결론5-FU연합DC-CIK세포능구현저억제위암BGC-823세포급기BGC-823-SP세포적생장,가위림상화료연합생물면역치료위암제공이론의거。
Objective To observe the inhibitory effect of 5-Fluorouracil(5-FU)combined with DC-CIK cells against gastric carcinoma cell line BGC-823 and the side population(SP) cells in vitro.Methods BGC-823-SP cells were isolated from gastric carci-noma cell line BGC-823 .And BGC-823 cells were set as group A and BGC-823-SP cells as group B .DC-CIK cells were generated by stimulating human peripheral blood mononuclear cells .Then MTT colorimetric assay was used to evaluate the killing rates of 5-FU,DC-CIK cells and their combination against the gastric carcinoma cells of group A and B .Results ①The killing rates of different concen-trations of 5-FU in group A were both higher than those in group B , i.e.the drug resistance to 5-FU of group B was stronger than that in group A.②DC-CIK cells could inhibit both BGC-823 cells and BGC-823-SP cells,and the inhibitory effects were gradually en-hanced with the increase of effect target ratio .At the same effect target ratio ,there were no statistical differences in DC-CIK cells of group A and group B ( P >0.05).③The killing rates of 5-FU combined with DC-CIK cells against the two groups gastric carcinoma cells were both higher than that of single treatment group .Conclusion 5-FU combined with DC-CIK cells can significantly inhibit the growth of gastric carcinoma cells and the SP cells ,and the results provide the theoretical basis for the clinical application of chemothera -py combined with immunotherapy in inhibition of gastric carcinoma .
