以合作猪为试验材料,采用PCR-SSCP检测442只合作猪SLA-DQB 基因第3外显子的多态性,克隆测序群体内变异产生的各等位基因序列,构建系统发育树以明确合作猪SLA-DQB 基因等位基因之间的遗传关系.结果表明:合作猪SLA-DQB 基因第3外显子中表现了7个基因型,其中有4个新等位基因,分别命名为B、C、D、E,序列分析中发现了11个核苷酸多态位点,这些多态位点主要由点突变形成,其中转换9个(占81.82%),颠换2个(占18.18%).表明合作猪SLA-DQB 基因第3外显子具有丰富的遗传多态性,群体中可能蕴藏着更多的抗性遗传资源.

Taking Hezuo pig as materal,SLA-DQB gene polymorphism of the 3rd exon from 442 pigs was detected based on PCR-SSCP method.Each allele produced by variation within population sequencing was measured to construct phylogenetic tree to ascertain the genetic relationship between alleles of SLA-DQB gene.The results indicated that the 3rd exon of SLA-DQB gene in Hezuo pig showed seven geno-types,including four new alleles,named B,C,D and E,respectively.The sequence analysis confirmed that there were 1 1 nucleotide polymorphism loci,which were formed by point mutation,including 9 convert (81. 82%)and 2 transversion (18.18%).In conclusion,the 3rd exon of Hezuo pig SLA-DQB gene have abun-dant genetic polymorphism,their group may contain the more resistant genetic resources.
