目的:探讨苦参碱对 HL-60白血病细胞增殖及凋亡的影响.方法:运用 MTT法和流式细胞术检测苦参碱对 HL-60白血病细胞的增殖抑制及促凋亡作用,应用实时定量 PCR技术检测 Bcl-2蛋白的表达.结果:随着苦参碱浓度的升高,各项指标与对照组相比较的差异均有统计学意义(P<0.01);HL-60细胞增殖明显受到抑制;细胞凋亡率明显增加,且当苦参碱浓度为2.0mg/ml时,细胞凋亡率达65.84%;2.0mg/ml 苦参碱作用72h 后的细胞,bcl-2蛋白的表达较对照组显著减少.结论:苦参碱具有对 HL60白血病细胞增殖抑制及促凋亡作用,bcl-2蛋白表达下调在此过程中可能发挥着重要的作用.

목적:탐토고삼감대 HL-60백혈병세포증식급조망적영향.방법:운용 MTT법화류식세포술검측고삼감대 HL-60백혈병세포적증식억제급촉조망작용,응용실시정량 PCR기술검측 Bcl-2단백적표체.결과:수착고삼감농도적승고,각항지표여대조조상비교적차이균유통계학의의(P<0.01);HL-60세포증식명현수도억제;세포조망솔명현증가,차당고삼감농도위2.0mg/ml시,세포조망솔체65.84%;2.0mg/ml 고삼감작용72h 후적세포,bcl-2단백적표체교대조조현저감소.결론:고삼감구유대 HL60백혈병세포증식억제급촉조망작용,bcl-2단백표체하조재차과정중가능발휘착중요적작용.
Objective:To investigate the effects of Matrine on the proliferation and apoptosis of HL-60 leukemia cells.Methods:Using the method of MTT and matrine flow cytometry was used to detect the inhibition on HL-60 leukemia cellproliferation and apoptosis,Bcl-2 protein expression was detected by real-time quantitative PCR technology.Result:With the increase of matrine concentration indexes,compared with control group,the differences were statistical y significant (P<0.01);the proG liferation of HL-60 cells was significantly inhibited;cellapoptosis rate increased significantly,and when the Matrine concentration is 2.0mg/ml,the apoptosis rate was 65.84%;2.0mg/ml matrine 72h cells,the expression of Bcl-2 protein was significantly reduced compared with the control group.Conclusion:Matrine has on the prolifG eration of HL60 leukemia cellinhibition and apoptosis,the expression of Bcl-2 protein may play an important role in the process of cut.
