Millettia Speciosa Champ%polysaccharide%antioxidant activity
建立了热水提取、乙醇沉淀、三氟乙酸除蛋白、Sephadex G-75凝胶过滤层析法,从牛大力分离纯化得到一种水溶性多糖,命名为MSP-1。通过红外光谱和离子色谱对其结构和单糖组成进行初步分析,结果表明MSP-1主要由鼠李糖、半乳糖、葡萄糖、甘露糖和果糖组成。同时对牛大力水提物、醇沉物和粗多糖进行体外抗氧化活性研究,结果表明,三者的抗脂质过氧化作用和对·OH自由基和DPPH·自由基清除能力:VC>牛大力水提物>牛大力醇沉物>牛大力粗多糖,并呈量效关系。

A water-soluble polysaccharide (MSP-1) was obtained from Millettia Speciosa Champ. by hot water extraction, ethanol precipitation, trifluoroacetic acid method removing proteins and gel permeation chromatography. The structure of MSP-1 was characterized by Ion Chromatography and FTIR spectroscopy. The results showed that MSP-1 was mainly composed of rhamnose, galactose, glucose, mannose and fructose. Additionally, Antioxidant activity in vitro of different extractions from Millettia Speciosa Champ were studied by testing the scavenging effects of ·OH, DPPH· and lipid peroxidantion. The results showed that three parts can effectively remove ·OH, DPPH·, lipid peroxidantion and was dose-effect relationship. The order of radical scavenging activity was as follow:water extraction>ethanol precipitation>crude polysaccharide.
