小球藻因其高的油脂含量和生长特性成为生物柴油研究的重要微藻资源之一,而研究表明油脂提取方法的不同通常直接关系到生物柴油的质量。研究了索氏提取、有机溶剂浸提两种油脂提取方法及不同细胞前处理方法对小球藻油脂提取率的影响,并比较了不同方法提取的油脂组成成分差异。结果表明,在不同的提取方法中,油脂得率、油脂中中长链脂肪酸(C14-C22)占总脂肪酸比例及脂肪酸提取纯度各不相同。其中,氯仿-甲醇浸提法是一种简便快捷高效的油脂提取方法,其油脂得率为(22.40±0.89)%,中长链脂肪酸(C14-C22)比例为85.84%;索氏提取法中无水乙醇作为提取溶剂获得的得率最高为(24.37±0.21)%,石油醚提取的油脂脂肪酸成分最多,达到10种。

Chlorella has become one of the important microalgae resources for biodiesel research because of its high fat content and growth characteristics,and there is a lot of research showing that the lipin extraction method is usually directly related to the quality of biodiesel.We investigate the Soxhlet extraction and organic solvent extraction of lipin extraction method and the influence of extraction rate of chlorella lipin extracted by different pre-cell approach methods, and compared the composition differences of lipin extracted by different methods.The results show that the different extraction methods result in different lipin yield ,different ratio of long-chain fatty acids (C14-C22)to total fatty acids and different extract purity of fatty acids.Among them,chloroform-methanol extraction method is a simple and efficient method of lipin extraction,with lipin yield of (22.40±0.89)%and 85.84%long chain fatty acids (C14-C22);In Soxhlet extraction,we get the highest yield rate of (24.37±0.21)%with Anhydrous alcohol as an extraction solvent,and we get up to 10 kinds of fatty acids with petroleum ether as an extraction solvent.
