目的:探讨苦碟子注射液(KDZ)的安全剂量及体外细胞毒性评价的可行性。方法:采用 MTT 法检测沈阳、通化两个厂家共12批次的KDZ对小鼠成纤维细胞(L929)的细胞毒性级别,并比较各两厂家之间的半数抑制浓度(IC50)差异是否存在统计学意义。结果:12批 KDZ 的给药浓度与 OD 值成显著的负相关,相关系数分别为-0.986和-0.924;12批KDZ原液对L929的细胞毒性级别均为4级,稀释64倍时毒性级别均为1级;两个厂家KDZ的IC50比较差异并无统计学意义(P>0.05)。结论:当KDZ稀释64倍时对L929细胞基本无毒,采用细胞评价方法对KDZ进行安全性评估是可行的。

Objective:To explore the feasibility of in vitro cytotoxicity and safe dose of Ku Die Zi injection(KDZ).Methods:MTT method was used to detect the cell toxicity level of 12 batches of KDZ of Shenyang Shuangding pharmaceutical and Tonghua Huaxia medicine on mouse fibroblast(L929).We compared the half inhibition concentration(IC50) of the two manufacturers whether there was significant difference.Results:The drug concentration of 12 batches of KDZ was negatively correlated with the OD value.The correlation coefficients were -0.986 and-0.924.The cytotoxic levels of L929 of 12 batches of KDZ solution were 4 classes,the toxicity levels were 1 when it was diluted 64 times.IC50 of two manufacturer KDZ had no significant difference(P>0.05).Conclusion:There is no toxic on L929 cell when KDZ is diluted 64 times,it is feasible for safety assessment of KDZ by using cell evaluation method.
