目的:评价龈下菌斑微生物表面抗原对人牙龈上皮细胞(HGECs)白细胞介素-1β(IL-1β)及基质金属蛋白酶-8(MMP-8)表达的影响。方法收集慢性牙周炎患者的龈下菌斑,经γ射线灭活制备表面抗原,按不同浓度稀释分为B、C、D、E、F组,A组为空白对照组。刺激人牙龈上皮细胞24 h后,采用ELISA检测细胞培养液中IL-1β和MMP-8含量,统计分析各组细胞因子的变化。结果龈下菌斑表面抗原刺激HGECs 24 h后,两种细胞因子表达的平均水平明显高于空白对照组(P<0.05)。IL-1β和MMP-8随抗原浓度的增加而升高,但达到峰值后下降。结论γ射线灭活可有效保留龈下菌斑微生物表面抗原;在细菌尚未侵入牙龈上皮细胞时,龈下菌斑表面抗原已促使细胞表达IL-1β和MMP-8,诱导牙周组织的炎症和免疫反应。

목적:평개간하균반미생물표면항원대인아간상피세포(HGECs)백세포개소-1β(IL-1β)급기질금속단백매-8(MMP-8)표체적영향。방법수집만성아주염환자적간하균반,경γ사선멸활제비표면항원,안불동농도희석분위B、C、D、E、F조,A조위공백대조조。자격인아간상피세포24 h후,채용ELISA검측세포배양액중IL-1β화MMP-8함량,통계분석각조세포인자적변화。결과간하균반표면항원자격HGECs 24 h후,량충세포인자표체적평균수평명현고우공백대조조(P<0.05)。IL-1β화MMP-8수항원농도적증가이승고,단체도봉치후하강。결론γ사선멸활가유효보류간하균반미생물표면항원;재세균상미침입아간상피세포시,간하균반표면항원이촉사세포표체IL-1β화MMP-8,유도아주조직적염증화면역반응。
Objective To elevate the effect of subgingival plaque surface antigen on human gingival epithelial cells secretion of cytokines interleukin-1β(IL-1β) and matrix metalloproteinase-8 (MMP-8). Methods Subgingival plaque were collected from chronic periodontitis and inactivated byγ-ray to form antigen. The study was divided into B, C, D, E, F groups with different antigen concentrations, and the A group as the control group. The IL-1βand MMP-8 level of HGECs stimulated 24 hours by antigen was detected by ELISA and analyzed statistically. Results Both cytokines expression of HGECs stimulated was significantly higher than the control group (P<0.05), and rised with increasing concentration, then downgraded after threshold. Conclusion Subgingival plaque microorganisms could be inactivated by γ-rays but retained antigenicity. Without bacterial invasion, subgingival plaque antigens promote human gingival epithelial cells express IL-1β and MMP-8, induced periodontal tissue inflammation and immune responses.
