目的:建立稳定有效的小鼠下肢缺血实验方法,为进一步活体研究下肢缺血性疾病提供动物模型。方法:电凝法制作裸鼠单侧下肢股动脉缺血模型,并经X线、Micro-CT、超声、磁共振成像、近红外激光及组织病理学全面评价模型。结果:缺血模型制作后,大体病理观察缺血侧下肢皮肤颜色苍白,X线及Micro-CT均显示股动脉起始处血管中断;彩色多普勒超声及近红外激光光谱仪证实缺血肌肉的血流量及血氧饱和度均显著下降;磁共振T2加权成像显示缺血肌肉水肿呈高信号,弥散加权成像显示缺血肌肉水分子弥散受限。组织病理学显示缺血肌纤维明显肿胀,肌间隙变窄。结论:本实验建立电凝法小鼠下肢缺血模型,并证明该模型成功构建,为进一步治疗下肢缺血性疾病及机制研究提供基础。

목적:건립은정유효적소서하지결혈실험방법,위진일보활체연구하지결혈성질병제공동물모형。방법:전응법제작라서단측하지고동맥결혈모형,병경X선、Micro-CT、초성、자공진성상、근홍외격광급조직병이학전면평개모형。결과:결혈모형제작후,대체병리관찰결혈측하지피부안색창백,X선급Micro-CT균현시고동맥기시처혈관중단;채색다보륵초성급근홍외격광광보의증실결혈기육적혈류량급혈양포화도균현저하강;자공진T2가권성상현시결혈기육수종정고신호,미산가권성상현시결혈기육수분자미산수한。조직병이학현시결혈기섬유명현종창,기간극변착。결론:본실험건립전응법소서하지결혈모형,병증명해모형성공구건,위진일보치료하지결혈성질병급궤제연구제공기출。
Objective:To develop hindlimb ischemia model of mice for studying the mechanism and therapeutic methods of lower-extremity peripheral arterial disease .Methods: After mice right hindlimb ischemia with electrocoagulation , the two hindlimb was evaluated by macropathology , X-ray and micro-CT, ultrasound , magnetic resonance imaging , near-infrared spectroscopy and histopathology .Results:After ischemia , the skin color in right hindlimb turned pale after surgery induced injury .X-ray and micro-CT images were obtained to assess the termination of the proximal origin of the right femoral artery .Compared to the left control hindlimb , a significant less level of blood flow and oxygen satuation was observed by ultrasound and near-infrared spectroscopy .In T2 weight imaging and diffusion weighted imaging , the presence of edema in the right ischemic muscle and the higher ADC value were observed compare to the control .Histological examination of the section of injured muscle showed that most of the muscle cells were round , and the extracellular space was tight .Conclusion:The mice hindlimb ischemic model was built successfully to further study the therapeutic methods of lower-extremity peripheral arterial disease .
