目的：检测凋亡抑制蛋白PED/PEA-15在胃癌中的表达，以探讨其与胃癌发生的联系及临床意义。方法收集山西医科大学第一附属医院2012年1月至12月间行手术切除且病理证实的胃癌手术标本，应用免疫组化方法分别检测40例胃癌组织、40例相应癌旁组织及40例相应肿瘤旁正常组织中凋亡抑制蛋白 PED/PEA-15的表达情况，实验结果根据阳性细胞的百分率和染色强度进行评价，三组间样本率的比较用行×列无序列联表χ2检验（检验水准α＝0.05），两组间样本率的比较用独立样本的χ2检验（检验水准α＝0.0167），PED/PEA-15表达与胃癌临床病理关系分析采用四格表资料χ2检验（检验水准α＝0.05）。结果凋亡抑制蛋白PED/PEA-15在胃癌组织中的阳性率为70%（28/40），在癌旁组织和正常组织中阳性率分别为25%（10/40）、13%（5/40），胃癌组织中 PED/PEA-15表达较癌旁组织、肿瘤旁正常组织显著增高（P＜0.05）。PED/PEA-15的表达与肿瘤细胞分化和临床分期相关（P＜0.05），与不同性别、年龄、肿瘤大小、转移无关（P均＞0.05）。结论 PED/PEA-15在胃癌组织中高表达，可能在胃癌发生发展过程中起着重要的作用，可作为判断人胃癌生物学行为的参考指标，且有望成为胃癌靶向基因治疗的新靶点。

Objective To detect the expression of inhibitor of apoptosis protein PED/PEA-15 in gastric cancer, and to investigate the relationship between ped/pea-15 and gastric cancer and its clinical significance. Methods Operation specimens of gastric cancer between January and December in 2012 were collected from the first affiliated hospital of Shan xi Medical University. All specimens had been proved by pathology. Immunohistochemical method was used to detect the expression of PED/PEA-15 in 40 cases of gastric cancer tissues, 40 cases of corresponding para carcinoma tissues and 40 cases of corresponding normal tissues adjacent to the tumor.The experimental result were evaluated according to the percentage of positive cells and the staining intensity. Row×columns unordered contingency table chi-square test was used in comparison of sample rate among three groups(test levelα=0.05).Chi-square test of independent samples was used in comparison of sample rate between the two groups(test level α=0.0167).The fourfold table chi-square test was used for analysis of relationship between expression of PED/PEA-15 and clinical pathology of gastric cancer(significance level α=0.05). Results The positive rate of the inhibitor of apoptosis protein PED/PEA-15 in gastric carcinoma, the tissue adjacent to carcinoma and normal tissues adjacent to the tumor was 70% (28/40), 25%(10/40) and 13%(5/40) respectively, The expression of PED/PEA-15 in gastric significantly increased compared with the adjacent cancer tissues and normal tissues adjacent to the tumor(P<0.05). The expression of PED/PEA-15 was associated with tumor cell differentiation and clinical stage (P<0.05), and wasn't associated with gender, age, tumor size, metastasis. (P>0.05). Conclusion The inhibitor of apoptosis protein PED/PEA-15 showed high expression in gastric carcinoma, which may play an important role in the occurrence and development of gastric carcinoma. Thus it may serve as a reference indicator for biological behaviors of gastric carcinoma and a new target for gene therapy.
