文中建立一种简单、快速测定猪肝中克伦特罗的液相色谱检测方法。鲜猪肝中克伦特罗在碱性条件下(pH-pKa>2),乙酸乙酯匀浆萃取并酸化乙酸乙酯有机基质后水反萃取,SCXSPE柱净化及氨化甲醇洗脱,氮吹近干后挥干,甲醇定容后测定。色谱分离采用Shisedo Capcell PAK SCX UG80150mm×4.6mm柱,乙腈、磷酸二氢钾为流动相,210nm波长处检测。该方法工作标准曲线的线性范围为185.6ng?ml-1~46.4μg?ml-1,相关系数r=0.99995,其检出限为0.15μg?ml-1,回收率为92.4~95.2%。该方法的准确性和精密度满足检测要求需要。

Objective a simple and quick method of determination of clenbuterol in pig liver by RP-HPLC is established in this paper. Method In the alkaline condition, clenbuterol in the fresh pig liver is homogenously extracted by acetic ether, and then acidiifcated, back extracted with water. Throught the puriifcation of SCXSPE column and the elution of ammonia methanol, nitrogen blowing to dry and diluting to a constant volume with methanol. In the chromatographic separation, Shisedo Capcell PAK SCX UG80 150mm×4.6mm column is served as isolated column, acetonitrile mixed with potassium dihydrogen phosphate solution is used as mobile phase and detection wavelength is 245 nm. Result Linear range of the standard curve of the method is 185.6ng?ml-1~46.4μg?ml-1 with the correlation coefifcient r=0.99995. The detection limit is 0.15μg?ml-1.The recovery rate of the method is 92.4~95.2%. The accuracy and precision of this method meet the detection requirements.
