目的:探讨大鼠肝细胞癌变过程中survivin和apollon的动态表达及意义。方法:以改良型间断饮用二乙基亚硝胺( diethylnitrosamine ,DEN)诱导大鼠原发性肝癌模型,观察大鼠肝脏在不同诱癌阶段的病理变化,采用免疫组织化学法和逆转录聚合酶链反应( RT-PCR)法检测大鼠HCC不同时期survivin和apollon的蛋白及mRNA表达。结果:改良型间断饮用DEN成功诱导了大鼠肝癌模型,诱癌结束时实验组大鼠自然死亡4只,死亡率5.88%(4/68)。实验组存活的16只大鼠15只形成肝癌,诱癌率93.75%(15/16)。免疫组化结果显示:survivin 和apollon 阳性表达率逐渐升高,各组间阳性率比较差异有统计学意义(P<0.05),survivin 和apollon 在肝癌组中表达呈正相关关系(r=0.515,p=0.003<0.05)。 RT-PCR检测结果显示,survivin mRNA和apollon mRNA表达,随着肝癌程度的加重而增加,与对照组肝组织比较,差异有统计学意义( P<0.05)。结论:survivin 、apollon在肝癌组织中呈高表达,提示二者在HCC的发生发展中起重要作用,可作为肝癌早期诊断的潜在标志物。

목적:탐토대서간세포암변과정중survivin화apollon적동태표체급의의。방법:이개량형간단음용이을기아초알( diethylnitrosamine ,DEN)유도대서원발성간암모형,관찰대서간장재불동유암계단적병리변화,채용면역조직화학법화역전록취합매련반응( RT-PCR)법검측대서HCC불동시기survivin화apollon적단백급mRNA표체。결과:개량형간단음용DEN성공유도료대서간암모형,유암결속시실험조대서자연사망4지,사망솔5.88%(4/68)。실험조존활적16지대서15지형성간암,유암솔93.75%(15/16)。면역조화결과현시:survivin 화apollon 양성표체솔축점승고,각조간양성솔비교차이유통계학의의(P<0.05),survivin 화apollon 재간암조중표체정정상관관계(r=0.515,p=0.003<0.05)。 RT-PCR검측결과현시,survivin mRNA화apollon mRNA표체,수착간암정도적가중이증가,여대조조간조직비교,차이유통계학의의( P<0.05)。결론:survivin 、apollon재간암조직중정고표체,제시이자재HCC적발생발전중기중요작용,가작위간암조기진단적잠재표지물。
Objective: To determine the dynamic expression and role of survivin and apollon during hepatocellular carcinogenesis ( HCC) in rats.Method:To establish modified DEN-induced primary hepato-carcinoma rat model and detect the protein and mRNA of survivin and apollon in the different pathologic peri-od during hepatocarcinogenesis by using immunohistochemistry and reverse transcriptase -polymerase chain reaction (RT-PCR).Result:The hepatoma model was induced by modified DEN , At the end, four rats died in experimental group, the mortality was 5.88%(4/68).In experimental group,in the sixteen survived rats, fifteen rat induced hepatocarcinoma ,the rate of hepatocarcinoma was 93.75%(15/16).The results of immu-nohistochemistry and RT-PCR shows that the proteins positive expression rate and expression intensity be-came higher and higher ,The difference between groups were statistically significant ( P<0.05) .The expres-sion of survivin was positively correlated with apollon expression in HCC .(r=0.515,p=0.003<0.05).Con-clusion:The expression of survivn and apollon protein is significantly high in human HCC indicating that survivin and apollon play an important role in hepatocarcinogenesis as early diagnostic stage of liver cancer .
