目的:观察吡格列酮对高脂血症大鼠主动脉基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)表达的影响。方法26只雄性SD大鼠随机分成普通饲料组( n =9)和高脂饲料组( n =17),分笼饲养,12周后检测空腹血脂水平,以确定高脂饲料组高脂血症大鼠造模是否成功;再将成模大鼠随机分成模型组( n =8)和吡格列酮组( n =9)。第13周起予吡格列酮组大鼠予吡格列酮混悬液连续灌胃4周,余2组用相应量的蒸馏水灌胃4周,平行检测2组大鼠血脂水平,免疫组化检测主动脉MMP-2、MMP-9表达,并用Image-Pro Plus 6.0图像分析系统分析染色结果。结果高脂饲料喂养12周大鼠总胆固醇( TC)、三酰甘油( TG)和低密度脂蛋白胆固醇( LDL-C)水平较对照组明显升高( P <0.05),表明造模成功。吡格列酮组主动脉MMP-2、MMP-9表达水平较之模型组明显降低( P <0.05)。结论吡格列酮能通过抑制主动脉MMP-2、MMP-9表达,有利于斑块的稳定性增加。

목적:관찰필격렬동대고지혈증대서주동맥기질금속단백매-2(MMP-2)、기질금속단백매-9(MMP-9)표체적영향。방법26지웅성SD대서수궤분성보통사료조( n =9)화고지사료조( n =17),분롱사양,12주후검측공복혈지수평,이학정고지사료조고지혈증대서조모시부성공;재장성모대서수궤분성모형조( n =8)화필격렬동조( n =9)。제13주기여필격렬동조대서여필격렬동혼현액련속관위4주,여2조용상응량적증류수관위4주,평행검측2조대서혈지수평,면역조화검측주동맥MMP-2、MMP-9표체,병용Image-Pro Plus 6.0도상분석계통분석염색결과。결과고지사료위양12주대서총담고순( TC)、삼선감유( TG)화저밀도지단백담고순( LDL-C)수평교대조조명현승고( P <0.05),표명조모성공。필격렬동조주동맥MMP-2、MMP-9표체수평교지모형조명현강저( P <0.05)。결론필격렬동능통과억제주동맥MMP-2、MMP-9표체,유리우반괴적은정성증가。
Objective To observe the effect of pioglitazone on the expression of MMP-2 and MMP-9 in aorta of rats with hyperlipidemia .Methods 26 male SD rats were randomly divided into normal diet control group ( n =9 ) and high-fat diet group ( n =16),and the rats were bred in respective cage .After 12 weeks,the fasting plasma lipid levels were detected to decide whether the animal models with hyperlipidemia in high fat diet group were successful or not .Then the model rats were redivided into model group ( n =8) and pioglitazone group ( n =9).From the 13th week,the rats in pioglitazone group were given pioglitazone (10mg· kg-1· d-1) by gavage for 4 weeks,however,the rats in the other two groups were given equivalent distilled water by gavage for 4 weeks,the blood fat levels in the two groups were detected ,the expression levels of MMP-2 and MMP-9 in aorta were detected by immunohistochemisty ,finally,the staining results were analyzed by Image Pro Plus 6.0 image analysis system .Results After 12-week feeding , the levels of total cholesterol ( TC ) , triglyceride ( TG ) and low-density lipoprotein cholesterol (LDL-C) in high fat diet group were significantly increased ,as compared with those in control group (P <0.05),which showed that the animal models were successfully established .The expression levels of MMP-2 and MMP-9 in aorta in pioglitazone group were significantly decreased ,as compared with those in model group ( P <0.05).Conclusion Pioglitazone can inhibit the expression of MMP-2 and MMP-9 in aorta,accordingly increase plaque stability .
