目的:研究二苯乙烯苷( THSG)对大鼠成骨细胞增殖、分化的影响。方法体外分离、培养SD大鼠成骨细胞,MTT法检测成骨细胞增殖活性;碱性磷酸酶( ALP)活性试剂盒检测成骨细胞ALP活性比;Elisa方法检测成骨细胞c-fos和c-jun蛋白表达水平。结果0.03~10 mg/ml的THSG干预成骨细胞24 h成骨细胞活性较对照组明显增强( P <0.05或<0.01);在二苯乙烯苷干预细胞后第3天时,二苯乙烯苷组成骨细胞ALP 活性较对照组明显增加( P <0.05或<0.01),成骨细胞c-fos和c-jun表达均较对照组明显增强( P <0.05或<0.01)。结论二苯乙烯苷能显著促进成骨细胞发育成熟。

목적:연구이분을희감( THSG)대대서성골세포증식、분화적영향。방법체외분리、배양SD대서성골세포,MTT법검측성골세포증식활성;감성린산매( ALP)활성시제합검측성골세포ALP활성비;Elisa방법검측성골세포c-fos화c-jun단백표체수평。결과0.03~10 mg/ml적THSG간예성골세포24 h성골세포활성교대조조명현증강( P <0.05혹<0.01);재이분을희감간예세포후제3천시,이분을희감조성골세포ALP 활성교대조조명현증가( P <0.05혹<0.01),성골세포c-fos화c-jun표체균교대조조명현증강( P <0.05혹<0.01)。결론이분을희감능현저촉진성골세포발육성숙。
Objective To investigate the effect of stilbene glycoside ( THSG) on proliferation and differentiation of rat’s osteoblasts.Methods The osteoblasts of SD rats were isolated and cultured in vitro .The cell proliferation activity was detected by MTT.The alkaline phosphatase ( ALP) activity was determined by alkaline phosphatase activity kit , and the expression levels of c-fos and c-jun in osteoblast were detected by ELISA .Results The osteoblast activities intervened by THSG at the concentration of 0.03~10mg/ml for 24h were significantly increased ,as compared with those in control group (P <0.05 or <0.01);the ALP activities of osteoblasts in THSG group after 3-day intervention were significantly increased, as compared with those in control group ( P <0.05 or <0.01);the expression levels of c-fos and c-jun in osteoblast were significantly higher than those in control group ( P <0.05 or <0.01) Conclusion Stilbene glycoside can promote the mature of osteoblasts obviously .
