目的探讨类固醇受体辅活化子( SRC)-3蛋白在严重烧伤后T淋巴细胞功能变化中的作用。方法健康SPF级雌性野生型( SRC-3+/+)小鼠、SRC-3基因敲除( SRC-3-/-)小鼠各10只,体质量约20 g,分为SRC-3+/+组( S组)和SRC-3-/-组(S-组),每组分为正常(N)和24h两个时相点,每个时相点各5只。制作15%~20%Ⅲo烧伤模型,按时脱臼处死小鼠,取股动脉血、脾脏和胸腺,并立即分离血浆和外周血、脾、胸腺淋巴细胞。测定血浆 IL-2、可溶性 IL-2受体(sIL-2R)表达和T淋巴细胞亚群的分布。结果两组正常小鼠血浆IL-2和sIL-2R水平没有显著差别,烧伤后24h均显著升高(P<0.01),但S-组小鼠血浆IL-2显著低于S组(P<0.01),而血浆sIL-2R显著高(P<0.01);正常情况下,S-组小鼠除脾脏CD8+高于S组小鼠(P<0.01)外,外周血、脾脏和胸腺CD3+、CD4+及CD4+ /CD8+比值均不同程度低于S组小鼠(P<0.05或P<0.01)。烧伤后24h,两组小鼠外周血、脾脏和胸腺CD3+、CD4+及CD4+ /CD8+比值均不同程度下降(P<0.05或P<0.01),CD8+显著增高(P<0.05或P<0.01),与S组相比,S-组外周血、脾脏和胸腺CD3+和CD4+降低更显著(P<0.01),CD4+ /CD8+比值差别均无统计学意义。结论 SRC-3蛋白可能与严重烧伤后T淋巴细胞免疫功能抑制有关,其蛋白缺失可引起T淋巴细胞亚群稳态的改变。

목적탐토류고순수체보활화자( SRC)-3단백재엄중소상후T림파세포공능변화중적작용。방법건강SPF급자성야생형( SRC-3+/+)소서、SRC-3기인고제( SRC-3-/-)소서각10지,체질량약20 g,분위SRC-3+/+조( S조)화SRC-3-/-조(S-조),매조분위정상(N)화24h량개시상점,매개시상점각5지。제작15%~20%Ⅲo소상모형,안시탈구처사소서,취고동맥혈、비장화흉선,병립즉분리혈장화외주혈、비、흉선림파세포。측정혈장 IL-2、가용성 IL-2수체(sIL-2R)표체화T림파세포아군적분포。결과량조정상소서혈장IL-2화sIL-2R수평몰유현저차별,소상후24h균현저승고(P<0.01),단S-조소서혈장IL-2현저저우S조(P<0.01),이혈장sIL-2R현저고(P<0.01);정상정황하,S-조소서제비장CD8+고우S조소서(P<0.01)외,외주혈、비장화흉선CD3+、CD4+급CD4+ /CD8+비치균불동정도저우S조소서(P<0.05혹P<0.01)。소상후24h,량조소서외주혈、비장화흉선CD3+、CD4+급CD4+ /CD8+비치균불동정도하강(P<0.05혹P<0.01),CD8+현저증고(P<0.05혹P<0.01),여S조상비,S-조외주혈、비장화흉선CD3+화CD4+강저경현저(P<0.01),CD4+ /CD8+비치차별균무통계학의의。결론 SRC-3단백가능여엄중소상후T림파세포면역공능억제유관,기단백결실가인기T림파세포아군은태적개변。
Objective To investigate the effect of steroid receptor coactivator ( SRC)-3 on T lymphocyte function after severe burns.Methods 10 female healthy SPF-grade SRC-3 +/ +mice and SRC-3-/-mice respectively, 20g and are named S group and S-group.Each group was divided into two phase points:normal (N) and 24h, which each of points had 5 mice.The distribution of T lymphocyte subsets of peripheral blood, spleen, thymus, and the levels of IL-2 and soluble IL-2 receptor ( sIL-2R) were deter-mined.Results There were no statistical significance between two groups about the levels of plasma IL-2 and sIL-2R under normal condition.They were both increased significantly (P<0.01) at 24h after burn in two groups, but the plasma IL-2 level was signifi-cantly lower in the S-group than that of S group (P<0.01) and the plasma sIL-2R content was markedly higher in the S-group than that of S group.Under normal condition, CD3 +, CD4 + and the ratio of CD4 + /CD8 + of peripheral blood, spleen and thymus were lower in the S-group than those of S group (P<0.05 or P<0.01), but spleen CD8 + was higher in the SRC-3-/-group than that of S group (P<0.01).After burn, the CD3 +, CD4 + and CD4 + /CD8 + ratio of peripheral blood, spleen and thymus were both de-creased (P<0.05 or P<0.01), the CD8 + were significantly increased (P<0.05 or P<0.01).Compared with the S group, the CD3 + and CD4 + of peripheral blood, spleen and thymus were decreased significantly ( P<0.01 ) , and there were no significant differences in the ratio of CD4 + /CD8 + in the S-group.Conclusion The SRC-3 protein might be related to the inhibition of T lym-phocyte immune function after severe burns, which its deletion can cause the distribution changes of T lymphocyte subsets.
