Klotho基因是与衰老有关的基因,编码分泌型和膜型两种蛋白质,在全身多系统表达。其表达产物可以作为糖苷酶水解TRPC-6发挥心肌保护作用,抑制心肌肥大并减少冠心病的发生;并可以作为循环因子抑制胰岛素样生长因子(IGF)发挥抗衰老作用,抑制转化生长因子(TGF)-β1发挥肾脏保护及抗肿瘤作用;作为FGF-23的共受体而对钙磷代谢产生影响。Klotho基因表达的调节也非常复杂,但因其各个系统的抗衰老作用,越来越受到人们的关注。

Klotho gene is associated with aging, encoding two kind of protein: membrane protein and secreted protein. It is expressed in multiple systems. The protein can act as aglycosidase and hydrolyze TRPV-6, in turn to inhibit myocardial hypertrophy and reduce the occurrence of coronary heart disease, it also act as circulating factor, inhibit insulin-like growth factor (IGF) and plays a role of anti-aging protein, inhibit transforming growth factor (TGF)-beta 1 to exert renal-protect anti-tumor effect;It modulate calcium/phosphorus metabolism as a co-receptor of FGF-23. The regulation of Klotho gene expression is very complex, but because of its anti-aging effect in various systems, more and more attention is paid to it.
