目的:观察菊花总黄酮对去势所致干眼症白兔泪腺凋亡相关基因蛋白Fas、FasL表达的影响,探讨菊花总黄酮治疗干眼症的作用机制。<br> 方法:将150只雄性日本大耳白兔随机分为正常组( A组)、假手术组(B组)、模型组(C组)、雄激素对照治疗组(D组)和菊花总黄酮治疗组(E组)。 C组、D组、E组行双侧去势术建立白兔干眼症模型。 E组白兔用菊花总黄酮灌胃治疗,D组用雄激素肌肉注射,A组、B组、C组用生理盐水灌胃。分别于治疗后1,3,5mo每组处死10只,取材用与相关指标检测。全部白兔行 SchirmerⅠ试验,并检测泪膜破裂时间,处死后取泪腺组织,采用免疫组织化学法检测泪腺组织中凋亡相关基因蛋白Fas、FasL的表达,并计数凋亡细胞数量。<br> 结果:E组SchirmerⅠ试验测量所得滤纸湿长明显高于C组(P<0.01),泪膜破裂时间明显长于C组(P<0.01)。 E组治疗1,3,5 mo后,泪腺导管及腺泡上皮细胞中Fas阳性表达的细胞数均明显低于C组,FasL阳性表达的细胞数均明显高于C组,细胞凋亡数量均明显低于C组(P<0.01)。结论:菊花总黄酮中主要成分为黄酮类物质,可显著抑制雄激素水平降低后白兔干眼症的发生,抑制泪腺细胞凋亡,维持泪腺基础分泌量和泪膜的稳定性。
目的:觀察菊花總黃酮對去勢所緻榦眼癥白兔淚腺凋亡相關基因蛋白Fas、FasL錶達的影響,探討菊花總黃酮治療榦眼癥的作用機製。<br> 方法:將150隻雄性日本大耳白兔隨機分為正常組( A組)、假手術組(B組)、模型組(C組)、雄激素對照治療組(D組)和菊花總黃酮治療組(E組)。 C組、D組、E組行雙側去勢術建立白兔榦眼癥模型。 E組白兔用菊花總黃酮灌胃治療,D組用雄激素肌肉註射,A組、B組、C組用生理鹽水灌胃。分彆于治療後1,3,5mo每組處死10隻,取材用與相關指標檢測。全部白兔行 SchirmerⅠ試驗,併檢測淚膜破裂時間,處死後取淚腺組織,採用免疫組織化學法檢測淚腺組織中凋亡相關基因蛋白Fas、FasL的錶達,併計數凋亡細胞數量。<br> 結果:E組SchirmerⅠ試驗測量所得濾紙濕長明顯高于C組(P<0.01),淚膜破裂時間明顯長于C組(P<0.01)。 E組治療1,3,5 mo後,淚腺導管及腺泡上皮細胞中Fas暘性錶達的細胞數均明顯低于C組,FasL暘性錶達的細胞數均明顯高于C組,細胞凋亡數量均明顯低于C組(P<0.01)。結論:菊花總黃酮中主要成分為黃酮類物質,可顯著抑製雄激素水平降低後白兔榦眼癥的髮生,抑製淚腺細胞凋亡,維持淚腺基礎分泌量和淚膜的穩定性。
목적:관찰국화총황동대거세소치간안증백토루선조망상관기인단백Fas、FasL표체적영향,탐토국화총황동치료간안증적작용궤제。<br> 방법:장150지웅성일본대이백토수궤분위정상조( A조)、가수술조(B조)、모형조(C조)、웅격소대조치료조(D조)화국화총황동치료조(E조)。 C조、D조、E조행쌍측거세술건립백토간안증모형。 E조백토용국화총황동관위치료,D조용웅격소기육주사,A조、B조、C조용생리염수관위。분별우치료후1,3,5mo매조처사10지,취재용여상관지표검측。전부백토행 SchirmerⅠ시험,병검측루막파렬시간,처사후취루선조직,채용면역조직화학법검측루선조직중조망상관기인단백Fas、FasL적표체,병계수조망세포수량。<br> 결과:E조SchirmerⅠ시험측량소득려지습장명현고우C조(P<0.01),루막파렬시간명현장우C조(P<0.01)。 E조치료1,3,5 mo후,루선도관급선포상피세포중Fas양성표체적세포수균명현저우C조,FasL양성표체적세포수균명현고우C조,세포조망수량균명현저우C조(P<0.01)。결론:국화총황동중주요성분위황동류물질,가현저억제웅격소수평강저후백토간안증적발생,억제루선세포조망,유지루선기출분비량화루막적은정성。
AIM: To evaluate the effects of total flavonoid of chrysanthemum on the expressions of Fas and FasL in male rabbits with dry eye, and to investigate the therapeutic effects of the total flavonoid of chrysanthemum on dry eye. <br> METHODS: Totally 150 male Japanses white rabbits were divided into blank group ( group A ) , sham -operated group ( group B ) , model group ( group C ) , androgen control treatment group (group D), and total flavonoid of chrysanthemum treatment group ( group E ) . The dry eye model was established with orchiectomy on group C, D and E. Rabbits in group E were treated with total flavonoid of chrysanthemum. Rabbits in group D were treated with androgen intramuscular injection. Rabbits in the group A, group B, group C was treated with normal saline. All rabbits were detected with Schirmer's Ⅰ test and tear break-up time (BUT). Fas, FasL were checked on immunohistochemistry. <br> RESULTS:The Schirmer's I test values of group E was significantly higher than that of group C ( P<0. 01 ) and the BUT value of group E was significantly longer than that of group C ( P<0. 01 ). The quantity of positive expression of Fas in glandular tube cell and acinar epithelial celland apoptosis cells of group E after treatment at 1, 3, 5mo were significantly lower than that of group C, cell population of the positive expression of FasL was obviously higher than that of group C (P<0. 01). <br> CONCLUSION:The main component of chrysanthemum is flavonoid, which could significantly inhibit happening of dry eye in rabbit after androgen level lowered and lacrimal gland apoptosis and keep basic tears secretory volume and tear film stability.
