建立脐橙中4种黄酮类功效成分及2种柠檬苦素类功效成分的RP-HPLC检测方法,同时测定其在脐橙中的含量。采用Agilent Eclipse XDB C18色谱柱(4.6×250 mm,5μm),流动相:梯度洗脱,A相:0.1%磷酸水;B相:乙腈,洗脱程序:23%B等度(0~5 min);23%~55%B线性变换(5 min~10 min);55%B等度(10 min~25 min);检测波长:波长切换技术,0~13 min:285 nm;13 min~25 min 210 nm;柱温:30℃;流速:1.0 mL/min。结果表明柚皮苷、橙皮苷、柚皮素、橙皮素、柠檬苦素、诺米林在1.7μg/mL~266.8μg/mL范围内线性关系良好,R2均≥0.9993,6种功效成分的平均加标回收率为94.6%~111.9%。建立的方法准确、可靠,可用于该6种功效成分在脐橙样品的含量测定。

To develop a novel quantitative method for simultaneous analysis of 6 functional components (Hesperidin, Naringin, Naringenin, Hesperetin, Limonin and Nomilin ) in navel orange by reversed-phase high performance liquid chromatography. The separation was performed on a Agilent Eclipse XDB C18 (4.6×250 mm, 5μm) and the column temperature was set at 30℃. The mobile phase was consisted of 0.1%phosphoric acid (A) and methanol (B), with a linear gradient elution at a flow rate of 1.0 mL/min. The gradient program was as flows:23%B isocratic (0-5 min);23%-55%B linear (5 min-10 min);55%B isocratic (10 min-25 min). And detection wavelength was 285 nm (0-13 min) and 210 nm(13 min-25 min). The correlation coefficients of the 6 calibration curves were all higher than 0.999 3. The scopes of the recovery rate was within the range of 94.6%-111.9 %, with RSDs no more than 2 %. The method is accurate and reliable, and can successfully analyze different navel orange samples.
