目的:建立测定人血浆中贝那普利浓度的反相高效液相色谱(HPLC)法。方法采用液-液萃取法分离纯化血浆样品,采用荧光检测器,检测波长为240 nm,以pH=2.2的0.02 mol/L(含3.0%三乙胺)磷酸盐缓冲液-甲醇(37:63)为流动相,色谱柱为Shim-Pack CLC-ODS柱(150 mm ×4.6 mm,3.5μm)。结果贝那普利色谱峰对称、不拖尾、无其他干扰,血药浓度线性范围为0.52~60.04μg/L ( r=0.9999),最低检测限为0.4μg/L,方法回收率为98.43%~100.68%,日内精密度 RSD 2.40%~4.20%,日间精密度 RSD 2.80%~4.80%。结论方法简便、准确,灵敏度高,选择性好,可用于贝那普利的血药浓度测定。

Objective To develop a HPLC method for quantifying the benazepril concentration in human plasma. Methods Sample plas-ma was isolated by the liquid-liquid extraction method and the UV detector was adopted with the detection wavelength of 240 nm. The mobile phase was phosphate buffer solution ( pH=2. 2, 0. 02 mol/L ) containing 3. 0% triethylamine-methanol ( 37 : 63 ); the chromato-graphic column was the Shim-Pack CLC-ODS column ( 150 mm × 4. 6 mm, 3. 5 μm ) . Results The chromatographic peak of benazepril was symmetric and non-trailing without other interference. The linear range of plasma benazepril concentration was 0. 52-60. 04μg/L ( r=0. 999 9 ) . The lowest detection limit of benazepril in plasma was 0. 4μg/L. The recovery rate was 98. 43% -100. 68%. The intra-day and inter-day RSD were 2. 40% -4. 20% and 2. 80% -4. 80% respectively. Conclusion The established method is simple, ac-curate, and highly sensitive, and can be used for detecting the benazepril concentration in plasma.
