本实验利用铜(Ⅱ)-1,10-菲啰啉-酸性铬蓝K三元络合物体系,结合巯基棉分离技术,建立双波长分光光度法测定饲料中铜含量的新方法。通过实验优化反应条件,并考察了共存离子对测定的干扰情况。结果表明:测量波长为548 nm,参比波长为609 nm,线性范围为0~0.6μg/mL,检测限为6.9μg/L,表观摩尔吸光系数为7.51×104 L/mol·cm。该方法用于测定饲料中铜含量,结果与原子吸收光谱法一致,相对标准偏差为0.95%~1.26%(n=5),样品加标回收率为97.6%~99.7%。

A new dual-wavelength spectrophotometric method was established for determination of copper content in feed based on the ternary complex system of Cu(Ⅱ)-1,10-phenanthroline -acid chrome blue K,combined with specia-tion technique of thiol cotton fiber.The reaction conditions for determination of Cu(Ⅱ) were optimized by experiments.The interferences from coexistent ions were also investigated.The results showed that the detection wavelength was at 548 nm and the reference wavelength was at 609 nm.The linear range of the determination was 0 ~0.6μg/mL,and the method de-tection limit of copper was 6.9 μg/L.The apparent molar absorption coefficient was 7.51 ×104 L/mol·cm.The method has been applied to the determination of copper in feed.The results obtained by this method agreed well with those obtained by atomic absorption spectrometry.RSD was in the range of 0.95% ~ 1.26%(n = 5),and the recovery of standard addition was in the range of 97.6%~99.7%.
