目的:为探讨PD早期诊断的生物标志物,疾病特异性蛋白质,应用蛋白质组学方法分析了鱼藤酮诱导慢性PD小鼠模型纹状体的蛋白质谱。方法构建鱼藤酮诱导慢性PD小鼠模型,行为学及免疫组化证明模型成功。提取小鼠脑纹状体蛋白质,运用双向电泳技术分离蛋白,Pdquest 8.0图形分析软件分析蛋白表达差异信息,用DE-PROMAL-DI-TOF质谱分析仪进行分析。结果与对照组相比,实验组共有32个蛋白点发生了变化:选其中1个有明显改变的蛋白:Centriolin(CP110)。结论本研究新鉴定出一个蛋白:CP110。以前研究表明该蛋白与神经退行性疾病有密切关系。推测此蛋白的显著改变可能与PD发病机制密切相关。

Objective:To application of proteomics research, nature can not only reveal the disease from the protein level, also contribute to the comprehensive study of Parkinson's disease (Pakinson 's disease, PD) pathological mechanism, establish the diagnosis criteria, found the drug target. Methods chronic PD mouse model induced by rotenone constructs, behavioral and immunohistochemical evidence model mice striatum, protein extraction, separation of proteins by two-dimensional gel electrophoresis, protein expression analysis software Pdquest 8 graphics, were analyzed by DE-PROMAL-DI-TOF mass spectrometry. Results compared with the control group, the experimental group a total of 32 protein spots were changed: choose one of 1 significant changes in protein: Centriolin (CP110). Conclusion this study identified a protein: CP110, former research indicates that there is a close relationship and neurodegenerative diseases. Presumably this protein significantly changes may be related to the pathogenesis of PD is closely related to, and lay the foundation for the study of Parkinson's disease.
