为了研究类球红细菌SOD的提取工艺,采用研磨法、超声波法、冻融辅助超声波法和酶法辅助超声波法提取类球红细菌超氧化物歧化酶,结果表明:酶法辅助超声波法是较优的提取方法。对类球红细菌SOD的提取工艺进行单因素和正交试验,得到较优的提取工艺参数为:超声波输出功率500W,超声波工作总时间15min,固液比1:15,酶添加量300μL,酶解温度40℃,酶解pH值7,酶解时间60min。在此较优提取工艺参数下,类球红细菌SOD活性达到385.7 U·mL-1,较优化前提高40%。

Extraction technique of SOD from Rhodobacter sphaeroides was studied by ultrasonic assisted with zymolysis. By comparision with grinding, ultrasonic and ultrasonic assisted with freezing-thawing, ultrasonic assisted with zymolysis was the optimum extraction method for superoxide dismutase (SOD) from Rhodobacter sphaeroides 3757. Results of both single factor experiment and orthogonal test showed that the optimum ultrasonic assisted with zymolysis extraction conditions of SOD were as follows: ultrasonic power 500W, ultrasonic total working time 15min, material/liquid ratio 1:15, amount of enzyme 300 μL, temperature of zymoIysis 40℃, pH of zymoIysis 7, time of zymoIysis 60 min. Under these optimum conditions, activities of SOD was 385.7 U·mL-1, and the extraction rate would be improved by 40%.
