目的 探讨一种新的建立裸鼠股骨骨肉瘤原位模型的方法.方法 体外连续传代筛选HOS细胞株,HOS细胞悬液闭合注射于股骨下端骨髓腔中.观察MNNG/HOS细胞在股骨骨肉瘤原位模型的成瘤特性.常规影像学检查以及肿瘤组织病理检查.结果 接种2周后平均肿瘤体积为(0.210±0.095) cm3,4周后平均肿瘤体积为(0.84±0.25) cm3.外观皮肤呈紫红色,静脉怒张.术后4周X线片可见股骨下段溶骨及成骨改变、肿瘤样骨形成.光镜下可见典型骨肉瘤病理特征,成瘤率为91.7%,免疫组织化学Stro-1阳性表达率为8%.结论 该方法建立模型操作简单,创伤小,模拟了人类骨肉瘤的临床发病和发展过程,提供了更加贴近人体的研究模型.

Objective To explore a new orthotopic transplantation method for establishment of human osteosarcoma in nude mice.Methods The cell suspensions of MNNG/HOS cells were transplantated into the distal femur of nude mice by using minimally invasive injection,and the formation of neoplasm was observed.One month after injection,the mice were subjected to X-ray examination and then killed.The tumor tissues were subjected to pathologic hematoxylin and eosin (HE) stain.The stro-1 expression was detected by using immunohistochemistry.Results At 2nd and 4th week after transplantation,the average tumor volume was (0.21 ±0.095) cm3 and (0.84±0.25) cm3,respectively.The skin showed fuchsia,and venous engorgement was seen.There existed osteogenic and osteolytic lesions in the distal femur,and the tumor-like bone.The typical characteristics of osteosarcoma of the tumor tissue were observed under a microscope.The tumor formation rate of transplantation was about 91.7%.Immunohistochemistry revealed the Stro-1 positive rate was 8%.Conclusion The orthotopic transplantation model of osteosarcoma in nude mice is established successfully.There are certain advantages in this orthotopic model such as simple operation,minimal invasion.It imitates the clinical onset and development of human osteosarcoma and offers a closer research model to human osteosarcoma.
