Severe Fever with Thrombocytopenia Syndrome Bunya Virus%Morphology%Atomic Force Microscopy
[目的]利用原子力显微镜对新布尼亚病毒进行观察。[方法]超速离心制备新布尼亚病毒HB29株,采用磷钨酸负染透射电子显微镜进行观察;制备新布尼亚病毒的原子力显微镜样片,在此基础上对病毒进行原子力显微镜观察,采用轻敲模式在大气常温下进行扫描成像。[结果]透射电子显微镜观察到新布尼亚病毒球形病毒粒子,并提供病毒二维图像,可见刺突结构;原子力显微镜则呈现了新布尼亚病毒三维图像,且可见病毒表面有凹凸不平的特征和边缘不平滑,同时获得表面粗糙度等可量化指标。两种方法最终得到相似的形态学结果。[结论]利用原子力显微镜首次观察到新布尼亚病毒的三维形态结构,与透射电镜观察相比,原子力显微镜是一种制样简单,观察直观的新型病毒形态学研究工具。
[目的]利用原子力顯微鏡對新佈尼亞病毒進行觀察。[方法]超速離心製備新佈尼亞病毒HB29株,採用燐鎢痠負染透射電子顯微鏡進行觀察;製備新佈尼亞病毒的原子力顯微鏡樣片,在此基礎上對病毒進行原子力顯微鏡觀察,採用輕敲模式在大氣常溫下進行掃描成像。[結果]透射電子顯微鏡觀察到新佈尼亞病毒毬形病毒粒子,併提供病毒二維圖像,可見刺突結構;原子力顯微鏡則呈現瞭新佈尼亞病毒三維圖像,且可見病毒錶麵有凹凸不平的特徵和邊緣不平滑,同時穫得錶麵粗糙度等可量化指標。兩種方法最終得到相似的形態學結果。[結論]利用原子力顯微鏡首次觀察到新佈尼亞病毒的三維形態結構,與透射電鏡觀察相比,原子力顯微鏡是一種製樣簡單,觀察直觀的新型病毒形態學研究工具。

The morphology of the severe fever with thrombocytopenia syndrome bunya virus(SFTSV) was investigated by the atomic force microscopy(AFM) . The ultracentrifuged SFTSV HB29 strains were negatively stained by phosphotungstic acid, and observed under the transmission electron microscopy (TEM). The SFTSV sample was prepared according to an AFM protocol, and then the SFTSV image was scanned with tapping mode at room temperature. The two-dimensional TEM image revealed a classical spike like sphere structure. The three-dimensional AFM image showed the characteristics and the morphology of the SFTSV strains. The SFTSV particle was an irregular sphere with an uneven edge. The measurements of the uneven edge were obtained in this study. This is the first time to have got the three-dimensional morphological structure of SFTSV by AFM. AFM is a new research tool for viral morphological observation with the advantages of simple preparing, 3D direct-viewing and visible interfaceing.
