目的建立灵芝孢子粉中灵芝多糖、总三萜皂苷的含量测定方法。方法采用紫外分光光度法,苯酚-浓硫酸比色法于波长492nm处测定灵芝多糖吸光度;以香草醛:高氯酸:冰乙酸(2:8:50)为显色剂,于波长555nm处测定总三萜皂苷的吸光度。结果多糖、总三萜皂苷的标准曲线的线性范围分别为0.0100~0.0300mg(r=0.9982);0.025~0.125mg(r=0.9996),多糖、总三萜皂苷的平均加样回收率分别为101.17%、100.74%,RSD分别为1.84%、1.29%。结论该方法简便、快速、准确,可用于灵芝孢子粉中多糖、总三萜皂苷的含量测定。

Objective To establish a method for content determination of total polysaccharides and triterpenoid saponin in the GLS.Methods UV Spectrophotometry,phenol-sulfuric acid;vanillin-glacial acetic acid-sulphuric acid-perchloric acid mixture at the wavelength of 492nm and 555nm absorbance.Results The linear range of total ginseng polysaccharides was 0.0100~0.0300mg(=0.9982);0.025~0.125mg(=0.9996).The recovery rate of the study was 101.17%、100.74%,RSD=1.84%、1.29%.Conclusion The results of simulation indicate that this method was accurate,reliable and reproducible.It was suggested that this method can be used to determine the content of total polysaccharides and triterpenoid saponin in the GLS.
