目的:建立测定人血浆中地西泮浓度的一阶导数分光光度法,为临床应用提供技术参考。方法采用一阶导数光谱法在240.0 nm波长处测定人血浆中地西泮浓度。结果地西泮在5.35~31.85μg/mL范围呈良好线性关系;其回归方程为D=1.241×10-3C+3.0706×10-4(r=0.9979);平均回收率为100.85%,RSD=1.88%(n=6)。结论该法操作简单,测定结果准确,适用于地西泮血药浓度的监测。

Objective To develop first order derivative spectrophotometry for the determination of Diazepam concentra-tion in human plasma, to provide the technical reference for its clinical application. Methods The first order derivative spectrophotometry was performed at the wavelength of 240.0 nm. Results Diazepam showed a good linear relationship within a ranger of 5.35-31.85 μg/mL, the regression equation was D=1.241í10-3C+3.0706í10-4 (r=0.9979); the average recovery was 100.85% and RSD=1.88%(n=6). Conclusion This method is simple to operate and accurate in the detec-tion results, and can be applied in the analysis of Diazepam in human plasma.
