目的:探讨三七理血脂肝方对大鼠非酒精性单纯性脂肪肝( non-alcoholic fatty liver , NAFL)的治疗作用,进而探讨其作用机制。方法:雄性 Wistar 大鼠36只,随机分成正常组( C)、模型组( M)、三七理血脂肝方组( Y),每组12只。 C组给予普通饲料, M和Y组予高脂饲料造模,4周后继续予高脂饲料同时Y组大鼠予相应药物干预4周。动物实验结束时,取3组大鼠肝脏组织用苏木精-伊红染色、油红O染色进行病理学观察,检测肝组织甘油三酯(TG)、总胆固醇(TC)含量。检测血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)的活性,以及血脂血糖指标TG、 TC、低密度脂蛋白胆固醇( LDL-C)、高密度脂蛋白胆固醇( HDL-C)、游离脂肪酸( FFA)、空腹血糖(FBG)。蛋白免疫印迹法检测肝组织细胞色素P450亚型2E1(cytochrome P4502E1, CYP2E1)和解耦联蛋白2(uncoupling protein-2, UCP2)的表达水平。结果: M组大鼠血清ALT和LDL-C 较C组显著升高, Y组中这两个指标较M组明显下调,并能降低血清FFA水平; M组大鼠肝组织出现弥漫性大泡性脂肪变性,肝组织 TG、 TC含量显著升高, Y组大鼠肝脏的脂肪变性从分布和程度上均有所减轻,肝脂含量降低。另外M组大鼠肝脏的CYP2E1的表达上调、 UCP2下调, Y组表达趋向正常水平。结论:三七理血脂肝方对大鼠NAFL治疗有效,方药的降脂理血功效可能通过抑制氧化应激、调节脂质代谢发挥降脂保肝作用。

목적:탐토삼칠리혈지간방대대서비주정성단순성지방간( non-alcoholic fatty liver , NAFL)적치료작용,진이탐토기작용궤제。방법:웅성 Wistar 대서36지,수궤분성정상조( C)、모형조( M)、삼칠리혈지간방조( Y),매조12지。 C조급여보통사료, M화Y조여고지사료조모,4주후계속여고지사료동시Y조대서여상응약물간예4주。동물실험결속시,취3조대서간장조직용소목정-이홍염색、유홍O염색진행병이학관찰,검측간조직감유삼지(TG)、총담고순(TC)함량。검측혈청병안산안기전이매(ALT)、천동안산안기전이매(AST)적활성,이급혈지혈당지표TG、 TC、저밀도지단백담고순( LDL-C)、고밀도지단백담고순( HDL-C)、유리지방산( FFA)、공복혈당(FBG)。단백면역인적법검측간조직세포색소P450아형2E1(cytochrome P4502E1, CYP2E1)화해우련단백2(uncoupling protein-2, UCP2)적표체수평。결과: M조대서혈청ALT화LDL-C 교C조현저승고, Y조중저량개지표교M조명현하조,병능강저혈청FFA수평; M조대서간조직출현미만성대포성지방변성,간조직 TG、 TC함량현저승고, Y조대서간장적지방변성종분포화정도상균유소감경,간지함량강저。령외M조대서간장적CYP2E1적표체상조、 UCP2하조, Y조표체추향정상수평。결론:삼칠리혈지간방대대서NAFL치료유효,방약적강지리혈공효가능통과억제양화응격、조절지질대사발휘강지보간작용。
Objective: To investigate the therapeutic efficacy of Sanqi Lixuezhigan recipe in the rat model of non -alcoholic fatty liver ( NAFL) , as well as the underlying mechanism.Methods: Thirty-six male Wistar rats were randomly divided into normal control group ( C) , model group ( M) and Sanqi Lixuezhigan recipe group ( Y) .The rats of control group was fed with normal diet, M group and Y group with high-fat diet for 8 weeks, and from the fifth week rats of Y group were also administrated with Sanqi Lixuezhigan recipe once a day.Serum alanine aminotransferase ( ALT) , aspartate aminotransferase ( AST) , triglycer-ides ( TG) , total cholesterol ( TC) , LDL-C, HDL-C, FFA and FBG were measured.Liver histopathology was observed through hematoxylin-eosin staining (HE stain) and oil red O staining.Liver TG and TC were also determined.In addition, the protein expression of liver CYP2E1 and UCP2 was detected by Western blot.Results: Serum ALT and LDL-C in the model rat increased obviously , while impaired again in the rat of Sanqi Lixuezhigan recipe treatment group.In the liver of M group , diffusing hepatic steatosis was observed in liver lobules , and the liver TG and TC level increased dramatically.Compared with model , less area and extent of steatosis were found in the rat liver of Y group.And serum FFA content also decreased in Y group.Western blot showed up-regulated expression of CYP2E1 and down-regulated expression of UCP2 in the liver of M group, while their expression tended to the normal levels in the liver tissue of treatment group.Conclusion: Sanqi Lixuezhigan recipe can effectively improve non-alcoholic fatty liver in the high fat diet induced rat model.The lipid-lowering and blood-regulating function for this complex recipe may enable it to ameliorate liver lipid accumulation and liver injury through inhibiting oxidative stress and improve lipid metabolism.
