以菠萝皮渣为原料,采用酸碱处理法从菠萝皮渣中提取菠萝皮渣纤维,主要对木聚糖酶酶解菠萝皮渣纤维制备低聚木糖的工艺进行研究。采用单因素试验法分析影响酶解效果的主要因素,利用正交试验优化酶解法的最佳工艺条件。结果表明,制备的菠萝皮渣纤维主要成分为粗纤维,约占90.09%,其中纤维素56.71%,半纤维素32.20%,酸不溶性木质素0.48%;木聚糖酶酶解菠萝皮渣粗纤维最优条件为反应温度50℃, pH值6,底物质量浓度60 mg/mL,酶用量1650 U,反应时间3 h,提取率为12.27%,低聚木糖平均聚合度DP为2.38。

The pineapple peel cellulose is extracted by acid-alkali method from pineapple peel, then the production of xylooligosaccharides from pineapple peel cellulose with xylanase is mainly studied. The main factors of enzymatic hydrolysis are analyzed by single-factor experiment, and the optimal condition of enzyme hydrolysis is determined by using the orthogonal experimental based on single-factor experiment. The results show that crude fiber accounted about 90.09% in pineapple peel cellulose. The main constituents of the crude fiber are cellulose that account for 56.71%, and hemicellulose account for 32.20%, lignin account for 0.48%. The optimal conditions of enzyme hydrolysis are action temperature 50 ℃, pH value 6, substrate concentration 60 mg/mL, enzyme dosage 1 650 U, and time 3 h. The extraction rate is 12.27% under these conditions, and the average polymerization degree (DP) of the xylooligosaccharide is 2.38.
