目的:观察橙皮素(HSP)对大鼠离体胸主动脉血管反应性的影响及其可能的机制。方法采用离体血管平滑肌张力实验方法,观察 HSP对 Sprague Dawley(SD)大鼠离体胸主动脉环的作用,并探讨其作用机制。结果 HSP 对氯化钾(KCl)和去氧肾上腺素(PE)预收缩的血管环具有浓度依赖性的舒张作用。去内皮后,HSP 不同浓度梯度(10-5 mol/L,3×10-5 mol/L,6×10-5 mol/L,10-4 mol/L)的舒张血管作用减弱。结论 HSP对KCl和PE预收缩大鼠离体胸主动脉环具有浓度依赖性舒张作用。

목적:관찰등피소(HSP)대대서리체흉주동맥혈관반응성적영향급기가능적궤제。방법채용리체혈관평활기장력실험방법,관찰 HSP대 Sprague Dawley(SD)대서리체흉주동맥배적작용,병탐토기작용궤제。결과 HSP 대록화갑(KCl)화거양신상선소(PE)예수축적혈관배구유농도의뢰성적서장작용。거내피후,HSP 불동농도제도(10-5 mol/L,3×10-5 mol/L,6×10-5 mol/L,10-4 mol/L)적서장혈관작용감약。결론 HSP대KCl화PE예수축대서리체흉주동맥배구유농도의뢰성서장작용。
Objective To investigate the effect of hesperetin (HSP)on the isolated thoracic aortic rings of rats and to explore under-lying the mechanism of the HSP.Methods The isolated thoracic aortic rings of male Sprague Dawley (SD)rats were mounted on the organ bath and tension was recorded.The effect of HSP on the isolated thoracic aortic rings with intact endothelium or denuded endo-thelium was precontracted by the potassium chloride (KCl,60 mmol/L)or phenylephrine (PE,10-6 mol/L),and the effects of HSP on the blood vessels reaction induced by various drugs were recorded with Powerlab biological signal assay system.Results HSP completely vasorelaxed the contractions induced by KCl and PE in a concentration dependent manner in endothelium intact and endo-thelium denuded rat aortic rings.When the contractions was induced by KCl and PE,L NAME (10-4 mol/L),Indo (10-5 mol/L)and BaCl2 (10-3 mol/L)had no significant inhibition on the vasorelaxant effects of HSP (P>0.05).When the contractions was induced by KCl,4 AP (10-3 mol/L)could weaken HSP diastolic function (P<0.05).When the contractions was induced by PE,4 AP (10-3 mol/L)could weaken HSP diastolic function (P<0.05).Conclusion HSP vasorelaxed the contractions induced by KCl and PE in a concentration dependent manner in endothelium intact and endothelium denuded rat thoracic aortic rings.Its mechanism might be related to the voltage dependent potassium channel (Kv).
