目的：探讨法舒地尔对大鼠肺动脉平滑肌细胞(PASMCs)收缩、迁移能力的影响。方法以体外培养的 PASMCs 为研究对象,分为正常培养对照组和不同浓度(10μmol/L,30μmol/L,50μmol/L)法舒地尔处理组,分别采用胶原凝胶实验及 Boyden Chamber实验方法对PASMCs的收缩和迁移能力进行分析。结果法舒地尔能够显著抑制PASMCs的收缩和迁移(P<0.01),并具有浓度依赖性。结论法舒地尔对PASMCs的收缩和迁移能力具有显著抑制作用,这可能是其治疗肺动脉高压的重要作用机制。
목적：탐토법서지이대대서폐동맥평활기세포(PASMCs)수축、천이능력적영향。방법이체외배양적 PASMCs 위연구대상,분위정상배양대조조화불동농도(10μmol/L,30μmol/L,50μmol/L)법서지이처리조,분별채용효원응효실험급 Boyden Chamber실험방법대PASMCs적수축화천이능력진행분석。결과법서지이능구현저억제PASMCs적수축화천이(P<0.01),병구유농도의뢰성。결론법서지이대PASMCs적수축화천이능력구유현저억제작용,저가능시기치료폐동맥고압적중요작용궤제。
Objectives To investigate the effects of fasudil on the contractility and migration of rat pulmonary artery smooth mus-cle cells (PASMCs).Methods PASMCs were incubated or not (controls)with different concentrations(10μmol/L,30μmol/L,50μmol/L)of fasudil.The contractility and migration of PASMCs were investigated by collagen gel contraction assay and boyden cham-ber assay,respectively.Results Fasudil effectively inhibited the contractility and migration of PASMCs (P<0.01),in a dose de-pendent manner.Conclusion Fasudil effectively inhibited the contractility and migration of PASMCs,which might serve as an impor-tant mechanism of its anti pulmonary arterial hypertension effects.
