介绍了蔗糖酶米氏常数测定的新方法,采用旋光仪测定蔗糖在蔗糖酶作用下分解为果糖和葡萄糖过程中溶液旋光度的变化,通过测定一系列不同初始浓度蔗糖溶液转化过程的动力学曲线,采用计算机软件处理数据,得到蔗糖酶的米氏常数值。采用成熟的物理化学实验技术,原理清晰,配合计算机数据拟合方法,实验方法简便、快速。用于该校材料、化学等专业本科物理化学实验教学,取得良好的教学效果。

A new experimental method for measuring Michaelis constant of invertase is described . The inversion of cane sucrose to glucose and fructose is catalyzed by a crude-extract from the the autolysis of a dried baker's yeast ,and the angle of rotation of polarized light passing through the solution is measured with a polarimeter .The kinetic curves of a series of solutions with varied initial concentration of cane sucrose are investigated .The data are treated with computer software and the Michaelis constant of invertase is calculated . By means of conventional experimental technology of physical chemistry and computer data fitting ,the enzyme-catalysis experiment becomes convenient and quick . The experiment is applied in the laboratory physical chemistry curriculum for the undergraduates majored in material and chemistry ,in which good effects of teaching are achieved .
