目的:构建硬皮病小鼠动物模型。方法用100μg/0.1ml 博来霉素每天皮下注射6周龄 BALB/ c 雄性小鼠背部皮肤3周,造模组与空白和 PBS 两组为对照组,各组为10只。取注射局部皮肤进行 HE 染色和 Massion 染色,镜下观察和测定各组皮肤厚度、胶原纤维含量,新鲜皮肤组织测定羟脯氨酸和胶原含量。结果3周后造模组较对照组,皮肤增厚变硬,弹性降低,真皮层明显增厚,胶原纤维明显增粗、膨大,毛囊明显减少,炎性细胞浸润,胶原纤维、羟脯氨酸和胶原含量明显升高。结论符合硬皮病皮肤病理改变,构建硬皮病小鼠动物模型成功。
目的:構建硬皮病小鼠動物模型。方法用100μg/0.1ml 博來黴素每天皮下註射6週齡 BALB/ c 雄性小鼠揹部皮膚3週,造模組與空白和 PBS 兩組為對照組,各組為10隻。取註射跼部皮膚進行 HE 染色和 Massion 染色,鏡下觀察和測定各組皮膚厚度、膠原纖維含量,新鮮皮膚組織測定羥脯氨痠和膠原含量。結果3週後造模組較對照組,皮膚增厚變硬,彈性降低,真皮層明顯增厚,膠原纖維明顯增粗、膨大,毛囊明顯減少,炎性細胞浸潤,膠原纖維、羥脯氨痠和膠原含量明顯升高。結論符閤硬皮病皮膚病理改變,構建硬皮病小鼠動物模型成功。
목적:구건경피병소서동물모형。방법용100μg/0.1ml 박래매소매천피하주사6주령 BALB/ c 웅성소서배부피부3주,조모조여공백화 PBS 량조위대조조,각조위10지。취주사국부피부진행 HE 염색화 Massion 염색,경하관찰화측정각조피부후도、효원섬유함량,신선피부조직측정간포안산화효원함량。결과3주후조모조교대조조,피부증후변경,탄성강저,진피층명현증후,효원섬유명현증조、팽대,모낭명현감소,염성세포침윤,효원섬유、간포안산화효원함량명현승고。결론부합경피병피부병리개변,구건경피병소서동물모형성공。
Objective To establish animal models with scleroderma in mice. Methods Bleomycin was subcutaneously injected into the back skin of male BALB/ c mice aged 6 weeks every day for 3 weeks at the dosage of 100μg/ 0. 1ml. Thirty male BALB/ c mice were divided into model-established group and blank control group and PBS control group, each group of ten mice. skin thickness and collagen fiber content of tissue of the injection of local skin were measured by HE staining and Massion stai-ning. Hydroxyproline and collagen content of fresh skin tissue were detected. Results Compared with the control group after 3 weeks, sclerotic skin in mice of model-established group was present thickening and hardening, the elasticity reduced, the dermis obvious thickening, hair follicles decreased significantly, collagen fibers increases obviously and enlargement, inflammatory cells infiltration and content of collagen fiber and hydroxyproline and collagen increased significantly. Conclusion Animal model in mice with scleroderma can be established successfully in BALB/ c mice after 3-week injection of bleomycin.
