目的:研究蠲痹颗粒对胶原诱导性小鼠关节炎Th17/Treg细胞平衡调控作用,揭示蠲痹颗粒治疗类风湿关节炎的作用机理。方法:建立牛II型胶原诱导性关节炎(CIA)模型,按临床评分将20只DBA/1小鼠分为CIA模型组和蠲痹颗粒组,每组10只。关节炎临床评分法观察CIA小鼠关节炎发病严重程度;流式细胞术检测脾淋巴细胞(CD4、CD8、B220)亚群比例、CD4+T细胞内Th17、Th1细胞因子及Treg细胞的表达。结果:蠲痹颗粒能显著降低CIA小鼠胶原关节炎的临床评分,对脾淋巴细胞(CD4、CD8、B220)亚群比例无明显影响;同时下调CIA小鼠CD4+T细胞胞内白细胞介素-17A的表达水平,在一定程度上促进Treg细胞的表达。结论:调控Th17细胞与Treg细胞平衡,下调促炎细胞因子白细胞介素-17A表达,可能是其治疗类风湿关节炎的作用机制之一。

목적:연구견비과립대효원유도성소서관절염Th17/Treg세포평형조공작용,게시견비과립치료류풍습관절염적작용궤리。방법:건립우II형효원유도성관절염(CIA)모형,안림상평분장20지DBA/1소서분위CIA모형조화견비과립조,매조10지。관절염림상평분법관찰CIA소서관절염발병엄중정도;류식세포술검측비림파세포(CD4、CD8、B220)아군비례、CD4+T세포내Th17、Th1세포인자급Treg세포적표체。결과:견비과립능현저강저CIA소서효원관절염적림상평분,대비림파세포(CD4、CD8、B220)아군비례무명현영향;동시하조CIA소서CD4+T세포포내백세포개소-17A적표체수평,재일정정도상촉진Treg세포적표체。결론:조공Th17세포여Treg세포평형,하조촉염세포인자백세포개소-17A표체,가능시기치료류풍습관절염적작용궤제지일。
Objective:To study the balanced regulation of Juanbi Granule on Th17/Treg cells in mice with collagen-induced arthritis in order to explore its mechanism in the treatment of rheumatoid arthritis. Methods:The arthritis models were induced by the bovine type II collagen.20 DBA/1 mice were divided equally into the CIA group and the Juanbi granule group according to the clinical score.The arthritis clinical score method was used to observe the seriousness of the onset of arthritis in CIA mice;the flow cytometry was used to detect the subpopulation of the splenic lymphocytes (CD4,CD8,B220),and the Th17,Th1 cytokines and the expression of Treg cells in CD4+T cells.Results:Juanbi Granule could significantly reduce the clinical score of CIA mice with collagen-induced arthritis,but had no significant effect on the subpopulation of the splenic lymphocytes (CD4,CD8,B220).Meanwhile,it down regulated the expression level of IL-17A in the CD4+T cells of CIA mice,promoting the expression of Treg cells in a certain extent. Conclusion:Regulating the balance of the Th17 cells and Treg cells and down-regulating the expression of the proinflammatory cytokine IL-17A may be one of its mechanisms in the treatment of rheumatoid arthritis.
