目的:研究内皮细胞脱落的内皮微粒(EMP)对体外培养的T淋巴细胞活化增殖和产生细胞因子的影响,探讨EMP在免疫应答中的作用。方法以人内皮细胞来源和SD大鼠内皮细胞来源的EMP干预BN大鼠淋巴细胞。采用流式细胞术检测T淋巴细胞(CD3+淋巴细胞)早期活化分子CD69的表达;采用酶联免疫吸附试验(ELISA)检测各组细胞培养液上清中相关细胞因子γ干扰素(IFN-γ)及白细胞介素2(IL-2)的表达水平。结果与对照组[加入等体积磷酸盐缓冲液(PBS)]相比,EMP各浓度组T淋巴细胞早期活化分子CD69和培养液上清IFN-γ的表达水平显著增高(P<0.05);中、高浓度的EMP可上调IL-2的表达,较对照组差异有统计学意义(P<0.05)。结论 EMP可刺激T淋巴细胞活化,诱导产生Th1型细胞因子,表明EMP可通过活化T淋巴细胞并产生一系列生物学效应,促进T细胞介导的细胞免疫应答。

목적:연구내피세포탈락적내피미립(EMP)대체외배양적T림파세포활화증식화산생세포인자적영향,탐토EMP재면역응답중적작용。방법이인내피세포래원화SD대서내피세포래원적EMP간예BN대서림파세포。채용류식세포술검측T림파세포(CD3+림파세포)조기활화분자CD69적표체;채용매련면역흡부시험(ELISA)검측각조세포배양액상청중상관세포인자γ간우소(IFN-γ)급백세포개소2(IL-2)적표체수평。결과여대조조[가입등체적린산염완충액(PBS)]상비,EMP각농도조T림파세포조기활화분자CD69화배양액상청IFN-γ적표체수평현저증고(P<0.05);중、고농도적EMP가상조IL-2적표체,교대조조차이유통계학의의(P<0.05)。결론 EMP가자격T림파세포활화,유도산생Th1형세포인자,표명EMP가통과활화T림파세포병산생일계렬생물학효응,촉진T세포개도적세포면역응답。
Objective To study the influence of vascular endothelial microparticles (EMP)on T lymphocyte activation in vitro and the cytokine production,and to study the roles of EMP on immune response.Methods Lymphocytes from BN rats were interfered by EMP derived from human endothelial cells and SD rat′s endothelial cells. The early activation CD69 expression of T lymphocytes (CD3 +lymphocytes)was measured by flow cytometry.Cytokines such as interferon-gamma (IFN-γ) and interleukin 2 (IL-2 ) were measured for expression by enzyme-linked immunosorbent assay (ELISA).Results Compared with those of control group [adding same volune of phosphate buffer (PBS)],the expressions of CD69 and IFN-γin culture supernatant of each EMP concentration group increased remarkably (P<0.05).Compared with those of control group,the expression of IL-2 increased remarkably in culture supernatant of intermediate and high EMP concentration groups with statistical significance (P<0.05).Conclusions EMP can induce T lymphocyte activation and Th1 cytokine production.EMP may play an important role in immune response through activating T lymphocytes.
