目的:研究单核细胞趋化蛋白-1(MCP-1)在慢性乙型病毒性肝炎(CHB)患者肝脏活检组织和血清的表达。方法采用酶联免疫吸附试验(ELISA)对124例CHB炎患者和20例对照组血清MCP-1水平进行检测,采用免疫组化和实时荧光定量PCR检测CHB患者肝组织中MCP-1蛋白和mRNA表达。结果对照组和CHB组血清中MCP-1水平分别是(265±44)ng/L和(157±80)ng/L ,CHB组血清MCP-1水平显著低于对照组(t=5.355,P<0.01)。CHB患者血清MCP-1水平与肝组织炎症分级(r=0.353,P=0.011)和纤维化分期(r=0.496,P<0.01)呈正相关。肝组织中MCP-1 mRNA水平与炎症分级和纤维化分期呈正相关( r=0.465,P<0.01;r=0.493,P<0.01)。肝组织中M C P-1蛋白表达与炎症分级和纤维化分期呈正相关( r=0.486, P=0.001;r=0.542,P<0.01)。结论 CHB患者血清中MCP-1水平下调,MCP-1表达水平与肝组织炎症分级、纤维化分期呈正相关。

목적:연구단핵세포추화단백-1(MCP-1)재만성을형병독성간염(CHB)환자간장활검조직화혈청적표체。방법채용매련면역흡부시험(ELISA)대124례CHB염환자화20례대조조혈청MCP-1수평진행검측,채용면역조화화실시형광정량PCR검측CHB환자간조직중MCP-1단백화mRNA표체。결과대조조화CHB조혈청중MCP-1수평분별시(265±44)ng/L화(157±80)ng/L ,CHB조혈청MCP-1수평현저저우대조조(t=5.355,P<0.01)。CHB환자혈청MCP-1수평여간조직염증분급(r=0.353,P=0.011)화섬유화분기(r=0.496,P<0.01)정정상관。간조직중MCP-1 mRNA수평여염증분급화섬유화분기정정상관( r=0.465,P<0.01;r=0.493,P<0.01)。간조직중M C P-1단백표체여염증분급화섬유화분기정정상관( r=0.486, P=0.001;r=0.542,P<0.01)。결론 CHB환자혈청중MCP-1수평하조,MCP-1표체수평여간조직염증분급、섬유화분기정정상관。
Objective To investigate the expression of monocyte chemoattractant protein -1(MCP-1) in serum and liver biopsies of patients with chronic hepatitis B (CHB) .Methods 124 patients with CHB and 20 normal controls were explored .MCP-1 was de-tected by ELISA ,real time PCR and immunologic histochemistry ,respectively .The concentrations of MCP-1 in serum were meas-ured by ELISA .Real-time PCR and immunohistochemistry were applied to detect mRNA and protein expression of MCP-1 in the liver biopsy ,respectively .Results The concentrations of MCP-1 were (157 ± 80)ng/L in serum of CHB patients and (265 ± 44) ng/L in control groups .There were significantly different between the two groups (t=5 .355 ,P<0 .01) .There was positive correla-tions between MCP-1 concentration in serum and grading of inflammation(r=0 .353 ,P=0 .011) ,staging of fibrosis(r=0 .496 ,P<0 .01) in CHB patients .And positive correlations were also found between mRNA levels of MCP-1 and grading of inflammation(r=0 .465 ,P<0 .01) ,staging of fibrosis(r=0 .493 ,P<0 .01) in liver biopsies of patients with CHB .At the same time positive correla-tions was observed between immunohistochemistry score and grading of inflammation (r=0 .486 ,P=0 .001) ,staging of fibrosis(r=0 .542 ,P<0 .01) .Conclusion Concentrations of MCP-1 are decreased in the sera of CHB patients .Positive correlation lies between the expression of MCP-1 and histological grading and staging of CHB .
