以美国红枫‘白兰地’(Acer rubrum‘Brandywine’)的带腋芽的茎段为外植体,进行了组织培养与快繁技术研究,结果表明:1)外植体的最佳取材时段为3-5月,最佳消毒方案是用3%的 NaClO 处理8 min ,污染率仅为2.22%,存活率达91.11%.2)启动培养中,3因素对其影响的主次顺序依次为培养基,IBA ,6-BA .在WPM ,MS ,1/2MS ,DKW 4种培养基中,WPM 对外植体启动培养影响显著高于其他3种,最佳培养基配方为WPM + IBA 0.5 mg/L+6-BA 2.0 mg/L .3)增殖培养的最佳培养基配方为WPM+0.002 mg/L TDZ+0.4 mg/L NAA ,增殖系数为1.37.4)壮苗培养的最佳培养基配方为WPM+0.5 mg/L NAA +1.0 mg/L GA3,苗的长势最好,茎粗且伸长明显,叶大且绿.5)根诱导培养中,4因素对其影响的主次顺序依次为6-BA ,GA3,培养基,IBA ,最佳培养基配方为MS+2.0 mg/L IBA+1.0 mg/L GA3,生根率高达93.3%.6)组培苗的炼苗移栽以先闭盖锻炼6 d ,后开盖锻炼2 d ,再移栽到沙与腐殖质等比混合的基质中,存活率最高,达86.7%.

With stems of axillary buds explants of A cer rubrum Brandy ,the tissue culture and rapid propa-gation procedure were studied .The results showed that (1) the best time to draw the material from the brandy was March to May ,and the optimal disinfection of explants was processed 8 minutes with 3% Na-ClO ,and the pollution rate was only 2 .22% ,the survival rate was 91 .11% .(2) The sequence of influence on Axillary bud initiation culture was as follows :medium ,IBA ,6-BA ,and the effect of WPM medium on primary culture was better than others ,the optimal medium was WPM +0 .5 mg/L IBA+2 .0 mg/L 6-BA . (3) The optimal medium of axillary bud was WPM +0 .001 mg/L TDZ+0 .4 mg/L NAA ,and the propaga-tion ratio was 1 .37 .(4) WPM+0 .5 mg/L NAA +1 .0 mg/L GA3 was the optimal medium to tube plant-let growing ,stem length were higher and stronger ,leaves were bigger and greener apparently than others under this condition .(5) The sequence of influence on root induction culture is as follows :6-BA ,GA3 ,me-dium ,IBA ,and the optimal medium is MS + 2 .0 mg/L IBA + 1 .0 mg/L GA3 ,and rooting rate reaches 93 .3% .(6) After 6 days’ closed cap and then 2 days’ opened cap to practice seedlings ,surviving of tube plantlet rates could reach 86 .7% w hen transplant into substrate w hich the ratio of sand and humus con-tents was 1∶1 .
