目的:探索一种程序简洁、易于操作的细胞株冻存与复苏的方法。方法采用常规方法体外培养肠癌细胞株SW480,胰酶消化后,收集细胞悬液,A组细胞采用传统冻存与复苏方法,B组采用改良的冻存与复苏方法,分别于2周、3个月、6个月后立即复苏冻存细胞进行培养,观察细胞的存活情况,并于1周内不同时间点用MTT法检测细胞增殖情况并进行比较。结果采用改良方法冻存与复苏的SW480细胞存活率及生长曲线与传统方法比较无显著差异,细胞贴壁率、形态学特点及增殖能力无明显变化。结论改良的细胞冻存体系以及快速复苏法是细胞株冻存与复苏的一种可行方法,且更加方便快捷。

Objective To explore a simple and easy method to cryopreserve and resuscitate SW480 cells. Methods Colon cancer SW480 cell line was cultured by conventional method in vitro and digested by trypsin, then the cell suspension was col-lected. In the experiment two groups were designed . Group A adopted traditional frozen storage as well as recovery method, that is, the solution was centrifuged , the supernatant was discarded , the cells were resuspended by the classical cryopreservation solution and cell suspension was subpackaged into three cryopreserved tubes. Group B adopted the improved method of cryopreservation and recovery, that is, without centrifugation, the cell suspension was directly mixed with improved freezing liquid and subpack-aged into three cryopreserved tubes. After two weeks, three months, six months, the two kinds of cells were recovered and cultured. The cell recovery situation and activity was detected at different growth point within one week with MTT method. Results There were no significant differences in the adhesions, morphological changes and proliferative capacity between SW480 cells treated by improved cryopreservation system and recovery methods and those by traditional methods. Conclusion The modified cell freezing system, foam wrapped method as well as the rapid resuscitation approach was feasible, more convenient and easier to operate.
