目的:探讨β-榄香烯对裸鼠直肠癌的放射增敏作用及其相关机制。方法采用细胞悬液接种法建立人直肠癌Colo320细胞株的裸鼠移植瘤模型,荷瘤后将裸鼠随机分为空白对照组,单纯药物组(40 mg/kgβ-榄香烯),单纯放疗组(4Gy),放疗+药物联合治疗组(40 mg/kgβ-榄香烯+4 Gy放射)4组,每组4~5只。观察肿瘤生长情况以及肿瘤组织病理形态,并利用Tunel法检测各组肿瘤细胞凋亡情况,利用免疫组化方法检测各组肿瘤细胞 Fas基因的表达情况。结果与空白对照组相比,β-榄香烯联合放射组肿瘤重量显著下降,显示出明显的抑制肿瘤生长的作用(P<0.05)。从病理形态上看,β-榄香烯联合放射组肿瘤组织出现大片坏死,坏死细胞的比例进一步增加。此外,在相同照射剂量下,与空白对照组和单纯放射组相比,β-榄香烯联合放射组裸鼠肿瘤体积明显变小,肿瘤细胞凋亡率和 Fas 基因阳性表达率明显增加(P<0.05)。结论β-榄香烯对人直肠癌Colo320细胞株的裸鼠移植瘤有放射增敏作用,此作用可能与 Fas基因介导的肿瘤细胞凋亡有关。
目的:探討β-欖香烯對裸鼠直腸癌的放射增敏作用及其相關機製。方法採用細胞懸液接種法建立人直腸癌Colo320細胞株的裸鼠移植瘤模型,荷瘤後將裸鼠隨機分為空白對照組,單純藥物組(40 mg/kgβ-欖香烯),單純放療組(4Gy),放療+藥物聯閤治療組(40 mg/kgβ-欖香烯+4 Gy放射)4組,每組4~5隻。觀察腫瘤生長情況以及腫瘤組織病理形態,併利用Tunel法檢測各組腫瘤細胞凋亡情況,利用免疫組化方法檢測各組腫瘤細胞 Fas基因的錶達情況。結果與空白對照組相比,β-欖香烯聯閤放射組腫瘤重量顯著下降,顯示齣明顯的抑製腫瘤生長的作用(P<0.05)。從病理形態上看,β-欖香烯聯閤放射組腫瘤組織齣現大片壞死,壞死細胞的比例進一步增加。此外,在相同照射劑量下,與空白對照組和單純放射組相比,β-欖香烯聯閤放射組裸鼠腫瘤體積明顯變小,腫瘤細胞凋亡率和 Fas 基因暘性錶達率明顯增加(P<0.05)。結論β-欖香烯對人直腸癌Colo320細胞株的裸鼠移植瘤有放射增敏作用,此作用可能與 Fas基因介導的腫瘤細胞凋亡有關。
목적:탐토β-람향희대라서직장암적방사증민작용급기상관궤제。방법채용세포현액접충법건립인직장암Colo320세포주적라서이식류모형,하류후장라서수궤분위공백대조조,단순약물조(40 mg/kgβ-람향희),단순방료조(4Gy),방료+약물연합치료조(40 mg/kgβ-람향희+4 Gy방사)4조,매조4~5지。관찰종류생장정황이급종류조직병리형태,병이용Tunel법검측각조종류세포조망정황,이용면역조화방법검측각조종류세포 Fas기인적표체정황。결과여공백대조조상비,β-람향희연합방사조종류중량현저하강,현시출명현적억제종류생장적작용(P<0.05)。종병리형태상간,β-람향희연합방사조종류조직출현대편배사,배사세포적비례진일보증가。차외,재상동조사제량하,여공백대조조화단순방사조상비,β-람향희연합방사조라서종류체적명현변소,종류세포조망솔화 Fas 기인양성표체솔명현증가(P<0.05)。결론β-람향희대인직장암Colo320세포주적라서이식류유방사증민작용,차작용가능여 Fas기인개도적종류세포조망유관。
Objective To explore the radiosensitivity ofβ-elemene on Colo320 cells transplanted tumor in nude mice and its molecular mechanisms.Methods Colo320 cells transplanted tumor model was established by cell suspension inoculation in nude mice.Then the transplanted mice with 0.8-1.0 cm3 tumor were randomly divided into 4 groups:control,β-elemene (40 mg/kg),radiation (4 Gy),andβ-elemene (40 mg/kg)+ radiation (4 Gy) groups,with four to five mice in each.Tumor weight and morphology were observed in each group.In addition,the apoptosis of tumor cells was measured by TUNEL and the expression of Fas gene was detected by immunohistochemistry.Results The nude mice transplanted tumor model was successfully established.Compared with that in control and simple radiation groups,tumor weight was significantly decreased inβ-elemene combined with irradiation group (P <0.05).At the same time,the apoptosis rate and the expression of Fas gene in tumor cells were significantly increased (P <0 .0 5 )inβ-elemene combined with irradiation group compared with control and simple radiation groups. Conclusion β-elemene could enhance the radiosensitivity of Colo320 cells transplanted tumor in nude mice probably by inducing Fas-mediated apoptosis of tumor cells.
