目的:研究田七口服液对SD大鼠桡骨骨折愈合过程中骨痂中骨形态发生蛋白2(BMP-2)和成纤维细胞生长因子2(FGF-2)表达的影响,探讨田七口服液促进骨折愈合的机制。方法:3月龄SD大鼠120只,造成右桡骨骨折后,随机分为田七口服液,阳性药物对照组(接骨七厘片组)、阴性对照组(生理盐水组),分别在实验的第3天、7天、14天、21天时,测量骨痂厚度,采用免疫组织化学方法检测BMP-2和FGF-2表达的变化。结果:田七口服液组和阳性药物对照组在骨折后14d BMP-2的表达达到峰值,两者间比较无显著性差异(P>0.05),与阴性对照组比较存在显著性差异(P<0.01)。田七口服液组在骨折后7d FGF-2的表达达到峰值,与阴性对照组比较差异显著(P<0.01),与阳性对照组比较无显著性差异(P>0.05)。结论:田七口服液能增加大鼠骨折端骨痂厚度,在骨愈合早期上调BMP-2和FGF-2的表达,促进骨折愈合。
目的:研究田七口服液對SD大鼠橈骨骨摺愈閤過程中骨痂中骨形態髮生蛋白2(BMP-2)和成纖維細胞生長因子2(FGF-2)錶達的影響,探討田七口服液促進骨摺愈閤的機製。方法:3月齡SD大鼠120隻,造成右橈骨骨摺後,隨機分為田七口服液,暘性藥物對照組(接骨七釐片組)、陰性對照組(生理鹽水組),分彆在實驗的第3天、7天、14天、21天時,測量骨痂厚度,採用免疫組織化學方法檢測BMP-2和FGF-2錶達的變化。結果:田七口服液組和暘性藥物對照組在骨摺後14d BMP-2的錶達達到峰值,兩者間比較無顯著性差異(P>0.05),與陰性對照組比較存在顯著性差異(P<0.01)。田七口服液組在骨摺後7d FGF-2的錶達達到峰值,與陰性對照組比較差異顯著(P<0.01),與暘性對照組比較無顯著性差異(P>0.05)。結論:田七口服液能增加大鼠骨摺耑骨痂厚度,在骨愈閤早期上調BMP-2和FGF-2的錶達,促進骨摺愈閤。
목적:연구전칠구복액대SD대서뇨골골절유합과정중골가중골형태발생단백2(BMP-2)화성섬유세포생장인자2(FGF-2)표체적영향,탐토전칠구복액촉진골절유합적궤제。방법:3월령SD대서120지,조성우뇨골골절후,수궤분위전칠구복액,양성약물대조조(접골칠전편조)、음성대조조(생리염수조),분별재실험적제3천、7천、14천、21천시,측량골가후도,채용면역조직화학방법검측BMP-2화FGF-2표체적변화。결과:전칠구복액조화양성약물대조조재골절후14d BMP-2적표체체도봉치,량자간비교무현저성차이(P>0.05),여음성대조조비교존재현저성차이(P<0.01)。전칠구복액조재골절후7d FGF-2적표체체도봉치,여음성대조조비교차이현저(P<0.01),여양성대조조비교무현저성차이(P>0.05)。결론:전칠구복액능증가대서골절단골가후도,재골유합조기상조BMP-2화FGF-2적표체,촉진골절유합。
Objective:To investigate the effects of Panax notoginseng on expression of BMP-2 and FGF-2 in bony callus in the process of radial fracture healing of SD rats’ fracture of tibia,and explored the mechanism of treating fracture with Panax notoginseng Methods:Radial fracture model was set up in SD rat, and 120 SD rats were divided randomly into the Panax notoginseng group,positive control group and negative control group. Selecting 3,7,14 and 21 days in the experiment, measuring the thickness of the callus and the immunohistochemistry method was adopted to detect the change of expression of BMP-2 and FGF-2.Results:Panax notoginseng group appeared as the peak of expression BMP-2 on l4th day after operation, compared with positive control group, significant differences were absent(P>0.05),but there were significant differences between Panax notoginseng group and negative control group(P<0.01). Panax notoginsen group could make expression of FGF-2 achieve peak value on 7th day after operation, compared to the positive control group:significant differences were absent (P>0.05), and significant differences were present comparing to the negative control group(P<0.01). Conclusion:In the early of bone repair, Panax notoginseng could promote fracture healing, through up-regulating expression of VEGF and FGF-2 at molecular level and increase the thickness of the callus.
