为高效制备玉米寡肽,在玉米蛋白粉 Alcalase酶水解的基础上,从 Protamex 酶、 Flavourzyme酶、Protex.7L酶、胰蛋白酶、木瓜蛋白酶等5种蛋白酶中筛选出Protex.7L酶为继续酶解玉米蛋白Alcalase酶水解物的最适用酶。通过单因素试验和响应面分析确定继续酶解制备玉米寡肽的最佳工艺参数为:加酶量1070 U · g -1、pH 8.0、温度44.0℃。在此条件下,体系的寡肽得率为(45.81±0.35)%,与理论预测值的相对误差在±1%以内,比单独应用Alcalase酶解提高25.04%。

Proteases ,Protamex ,Flavourzyme ,Protex .7L ,trypsin and papain were tested for the hydrolysis of Alcalase-hydrolyzed corn gluten meal for oligo-peptide preparation . From the single factor experiment using the response surface methodology ,the optimized conditions for the enzymatic hydrolysis were found to be Protex.7 L usage at a rate of 1 070 U · g -1 ,pH at 9.0 ,and temperature at 44.0℃ .The oligo-peptide yield was up to (45.81 ± 0.35)% ,reaching the predicted maximum with a relative error of less than ± 1% .That was 25.04% higher than that when the hydrolysis was carried out using Alcalase alone .
