目的:观察大鼠骨髓间充质干细胞(mesenchymal stem cells, MSCs)移植对正己烷致周围神经病大鼠神经电生理的影响。方法 SD大鼠54只随机分为对照组、低剂量染毒组和高剂量染毒组,低剂量组和高剂量组均用2,5-己二酮(2,5-HD),分别以200 mg/kg和400 mg/kg的剂量进行腹腔注射染毒,对照组用生理盐水进行腹腔注射。染毒6周后,以上各组均随机分为2组,分别任取1组大鼠尾静脉移植MSCs,每只大鼠移植1×106个MSCs。染毒及MSCs移植过程中,观察大鼠的一般状态和步态变化,并用肌电图与诱发电位仪记录大鼠尾神经传导潜伏期和神经传导速度。结果大鼠染毒6周时,低、高剂量组的步态评分分别升高至2.87±0.28和3.78±0.24、神经传导潜伏期分别延长为(5.15±0.29) ms和(6.75±0.35) ms、神经传导速度分别减低为(13.20±0.51) m/s和(8.00±0.43)m/s。 MSCs移植后,随移植时间的延长,模型大鼠步态评分下降,神经传导潜伏期和神经传导速度逐渐恢复。移植5周后,低、高剂量组的神经传导潜伏期分别恢复21.63%和16.08%,神经传导速度分别提高13.38%和38.71%,与未移植MSCs的对照组相比,差异有显著性意义(P<0.05)。结论 MSCs移植能明显改善正己烷致周围神经病大鼠的行为学和神经电生理。

목적:관찰대서골수간충질간세포(mesenchymal stem cells, MSCs)이식대정기완치주위신경병대서신경전생리적영향。방법 SD대서54지수궤분위대조조、저제량염독조화고제량염독조,저제량조화고제량조균용2,5-기이동(2,5-HD),분별이200 mg/kg화400 mg/kg적제량진행복강주사염독,대조조용생리염수진행복강주사。염독6주후,이상각조균수궤분위2조,분별임취1조대서미정맥이식MSCs,매지대서이식1×106개MSCs。염독급MSCs이식과정중,관찰대서적일반상태화보태변화,병용기전도여유발전위의기록대서미신경전도잠복기화신경전도속도。결과대서염독6주시,저、고제량조적보태평분분별승고지2.87±0.28화3.78±0.24、신경전도잠복기분별연장위(5.15±0.29) ms화(6.75±0.35) ms、신경전도속도분별감저위(13.20±0.51) m/s화(8.00±0.43)m/s。 MSCs이식후,수이식시간적연장,모형대서보태평분하강,신경전도잠복기화신경전도속도축점회복。이식5주후,저、고제량조적신경전도잠복기분별회복21.63%화16.08%,신경전도속도분별제고13.38%화38.71%,여미이식MSCs적대조조상비,차이유현저성의의(P<0.05)。결론 MSCs이식능명현개선정기완치주위신경병대서적행위학화신경전생리。
Objective To evaluate the effects of mesenchymal stem cells transplantation on neurophysiology of rats with pe -ripheral neuropathy induced by n -Hexan.Methods Total 54 rats were randomly divided into 3 groups, of which the rats were given at 0mg/kg, 200 mg/kg and 400 mg/kg 2,5-HD i.p.After 6 weeks, the rats injected with 2,5-HD were di-vided into two groups , and the rats in one group were transplanted with MSCs at 1 ×106 .The gait, distal latency and motor nerve conduction velocity were observed .Results At 6 weeks, the gait score was 3.78 ±0.24, the distal latency was (6.75 ±0.35)ms and motor nerve conduction velocity was (8.00 ±0.43)m/s in the rats of 400 mg/kg group.And these data in 200 mg/kg group were 2.87 ±0.28, 5.15 ±0.29 and 13.20 ±0.51 respectively.After MSCs transplantation, the recovered gait , decreased distal latency and increased motor nerve conduction velocity were observed in transplantation groups.At 5 weeks after MSCs transplantation , the gait score significantly decreased to 28.63%, the distal latency was re-covered to 21.63%and 16.08%, and the increased motor nerve conduction velocities were 13.38%and 38.71%in 200 mg/kg group and 400 mg/kg group, respectively.Conclusion MSCs transplantation can improve neurobehavior and neuro-physiology in peripheral neuropathy rats induced by n -Hexan.
