目的:观察山楂原花青素对缺血性脑中风大鼠血液流变学指标以及血脑屏障上P-gp表达的影响,探讨其可能的分子作用机制。方法采用线栓法制备缺血性脑中风大鼠模型,分为假手术组、模型组、山楂原花青素低、中、高剂量组,采用全自动血液流变学检测仪检测大鼠全血粘度、血小板最大聚集率、凝血酶时间和凝血酶原时间,采用高效液相色谱分析仪检测血浆NO水平,采用免疫组化法观察大鼠脑皮质缺血区P-gp的表达。结果山楂原花青素对缺血性脑中风大鼠全血粘度有降低作用,对血小板最大聚集率有抑制作用,对血浆NO有升高作用,与模型组比较有显著性差异(P<0.01),而对凝血酶时间和凝血酶原时间影响不大,差异无统计学意义(P>0.05),山楂原花青素对大鼠脑皮质P-gp的表达有显著抑制作用。结论山楂原花青素对缺血性脑中风大鼠有保护作用。

목적:관찰산사원화청소대결혈성뇌중풍대서혈액류변학지표이급혈뇌병장상P-gp표체적영향,탐토기가능적분자작용궤제。방법채용선전법제비결혈성뇌중풍대서모형,분위가수술조、모형조、산사원화청소저、중、고제량조,채용전자동혈액류변학검측의검측대서전혈점도、혈소판최대취집솔、응혈매시간화응혈매원시간,채용고효액상색보분석의검측혈장NO수평,채용면역조화법관찰대서뇌피질결혈구P-gp적표체。결과산사원화청소대결혈성뇌중풍대서전혈점도유강저작용,대혈소판최대취집솔유억제작용,대혈장NO유승고작용,여모형조비교유현저성차이(P<0.01),이대응혈매시간화응혈매원시간영향불대,차이무통계학의의(P>0.05),산사원화청소대대서뇌피질P-gp적표체유현저억제작용。결론산사원화청소대결혈성뇌중풍대서유보호작용。
Objective To observe the effect of hawthorn procyanidins(HPC) on blood rheology indexes and expression of P-glycoprotein (P-gp) in blood brain barrier in ischemic stroke rats ,and to explore its possible molecular mechanism .Methods The ischemic stroke rat model was prepared by thread approach and all rats were randomly divided into sham-operation group , model group and low-dose ,mid-dose ,high-dose of HPC .The whole blood viscosity (WBV) ,platelet maximum aggregation rate (PMAR) ,thrombin time(TT) and prothrombin time(PT) were detected by automatic rheometer .The level of plasma NO was detected by high performance liquid chromatograph .The expression of P-gp in ischemic cortex of rats was detected by immuno-histochemistry assay .Results HPC have significantly reduced the blood viscosity ,inhibited the platelet maximum aggregation rate ,and elevated the level of plasma NO when compare with model group (P<0.01) .But the effect on the thrombin and pro-thrombin time were not significantly different to the model group (P>0.05) .On the other hands ,HPC has significantly inhibi-tory effect on ischemic cortex of P-gp expression in ischemic stroke rats .Conclusion HPC have protective effect on ischemic stroke rats .
