目的:研究 PI3K 和 caspase-3在食管鳞癌组织(esophageal squamous cell carcinoma,ESCC)中的表达及其与食管肿瘤发生、发展与转移的关系。方法应用免疫组织化学链霉菌抗生物素蛋白过氧化物酶链结(SP)法检测24例正常食管黏膜、94例食管鳞癌组织中 PI3K 和 caspase-3的表达情况,并分析其与临床病理特征之间的关系。结果 PI3K 在食管鳞癌中的表达高于正常食管黏膜(P <0.01);PI3K 蛋白在高中分化鳞癌中表达低于低分化鳞癌,随着浸润深度和临床分期的增加,其表达阳性率增高,在淋巴结转移阳性组的表达高于阴性组(92.0% vs 47.7%, P <0.01);而 caspase-3阳性表达与 PI3K 相反,高中分化鳞癌中表达高于低分化鳞癌,随着浸润深度和临床分期的增加,其表达阳性率降低,在淋巴结转移阳性组的表达低于阴性组(38.0% vs 68.2%,P <0.01);PI3K 与 caspase-3表达有关联(r =-0.230,P <0.05)。结论 PI3K 高表达和 caspase-3低表达与食管鳞癌发生发展及侵袭转移存在协同作用,是反映食管鳞癌生物学行为的重要指标。

목적:연구 PI3K 화 caspase-3재식관린암조직(esophageal squamous cell carcinoma,ESCC)중적표체급기여식관종류발생、발전여전이적관계。방법응용면역조직화학련매균항생물소단백과양화물매련결(SP)법검측24례정상식관점막、94례식관린암조직중 PI3K 화 caspase-3적표체정황,병분석기여림상병리특정지간적관계。결과 PI3K 재식관린암중적표체고우정상식관점막(P <0.01);PI3K 단백재고중분화린암중표체저우저분화린암,수착침윤심도화림상분기적증가,기표체양성솔증고,재림파결전이양성조적표체고우음성조(92.0% vs 47.7%, P <0.01);이 caspase-3양성표체여 PI3K 상반,고중분화린암중표체고우저분화린암,수착침윤심도화림상분기적증가,기표체양성솔강저,재림파결전이양성조적표체저우음성조(38.0% vs 68.2%,P <0.01);PI3K 여 caspase-3표체유관련(r =-0.230,P <0.05)。결론 PI3K 고표체화 caspase-3저표체여식관린암발생발전급침습전이존재협동작용,시반영식관린암생물학행위적중요지표。
Objective To study the expression of PI3K and caspase-3 in esophageal carcinoma for the relationship between the expression of PI3K and caspase-3 as well as carcinogenesis or progression of esophageal carcinoma.Methods PI3K and caspase-3 expression were detected in 24 normal esophageal mucosa,94 primary tumor tissues with SP immunohistochemical method.Results The expression of PI3K in esophageal carcinoma was higher than that of normal mucosa epithelium(P <0.01).PI3K expression in higher-differentiated group was lower than that in low-differentiated group.With invasive depth and clinical staging aged,its expression became higher,so did it with metastasis of lymph node (92.0% vs 47.7%,P < 0.01 ).But the positive expression rate of caspase-3 was on the contrary.In higher-differentiated group,its positive expression rate was higher than in low-differentiated group.With invasive depth and clinical staging aged,its expression tended lower.In carcinoma with lymph node metastasis,it showed lower expression than in that of lymph node non-metastasis (38.0% vs 68.2%,P < 0.01 ).There was correlation between PI3K and caspase-3 expression in this study (r = -0.230,P < 0.05 ).Conclusion PI3K and caspase-3 are over expressed in esophageal carcinoma.They play important roles in carcinogenesis and progression of esophageal carcinoma and can be used as valuable biomarkers to evaluate biological characteristics in esophageal carcinoma.
