目的建立模拟外周血中筛查肿瘤细胞的方法，作为肿瘤诊疗的辅助手段，协助判断治疗效果以及预后推断。方法抗凝外周血，分离有核细胞，吖啶橙染色，荧光显微镜观察肿瘤细胞。结果因吖啶橙高度异染性，使肿瘤细胞变为彩色，加之肿瘤细胞特有的形态学特点，使得肿瘤细胞易于与正常细胞分辨开来。该方法在肿瘤细胞系、肿瘤组织印片、混有肿瘤细胞的外周血中应用都能使肿瘤细胞得到很好呈现，且特异性和敏感性都理想。在部分肾癌患者中验证，可检测到循环肿瘤细胞。结论吖啶橙染色法可用于血液中肿瘤细胞的染色，特异性、敏感性均较好，可用于临床协助判断治疗效果以及预后判定，为患者制定更合理的治疗方案。

Objective To establish the method of screening the circulating tumor cells to facilitate the diagnosis and prognosis prediction of cancer. Methods Isolate nucleated cells from anticoagulant peripheral blood,stain with acridine orange,and observe cells under fluorescence microscopy. Re-sults Acridine can dye the tumor cells,and show the morphologic characteristics of tumor cells un-der fluorescence microscopy.Acridine orange stain can make it easy to differentiate the tumor cells from normal cells. Conclusions Acridine orange staining method can be useful for circulating tumor cells screening.This method has an excellent specificity and sensitivity,which could be com-plementary for clinical diagnosis and treatment of tumor for better therapy.
