Weixuening%HPLC%fingerprints
目的:建立维血宁的高效液相色谱指纹图谱。方法采用HPLC法,以虎杖苷、大黄素-8-O-β-D葡萄糖苷、大黄素甲醚-8-O-β-D葡萄糖、大黄素和大黄素甲醚为内参照物,色谱柱为AccurasilC18(250mm×4.6mm,5μm),流动相为乙腈-1mL·L-1磷酸水(梯度洗脱),检测波长为280nm,流速为1.0mL·min-1,柱温为30℃,分析时间为90min。结果在HPLC指纹图谱中确定了5个主要共有峰,建立了维血宁的对照图谱。相似度比较结果显示,10批维血宁的对照图谱相似度均大于0.900。结论采用HPLC 方法建立维血宁指纹图谱,方法稳定、可靠、简便,色谱图展现的各峰分离度较好,特征明显,可作为维血宁真伪鉴别标准,为维血宁的质量评价提供参考依据。

Objective To establish a fingerprint analytical method of Weixuening .Methods By using polydatin ,emodin-8-O-β-D-glu-coside ,physcionas-8-O-β-D-glycosidase ,emodin and physcionas as reference substances ,the chromatographic separations were per-formed on an Accurasil C18 (250 mm × 4 .6 mm ,5 μm) column ,the column temperature was maintained at 30 ℃ ,a linear elution of 1 mL · L -1 phosphoric acid aqueous solution-acetonitrile was adopted at a flow rate of 1 mL · min-1 ,the UV detection wavelength was 280 nm and the analysis time was 90 min .Results There were five main common peaks in the fingerprint .The similarity of samples was all over 0 .900 .Conclusion The HPLC method for establishing the fingerprint was stable ,reliable ,and simple ,and the obtained chromatograms showed good separation and obvious peak characteristics ,which can be used as the standard for the identi-fication of decoction pieces in Weixuening .
